Abstract


Research background hypertrophic scars and keloids are two common clinical pathological scar, often caused by locally varying degrees of deformity and dysfunction. Due to the reasons for the formation of hypertrophic scars and keloids, lack of awareness of the mechanism, although more treatment options, clinical efficacy is not satisfactory. To date, not a drug or technology to ensure the success of the prevention and treatment of hypertrophic scars and keloids. Curcumin from Zingiberaceae Turmeric is a perennial herb turmeric extract natural pigment, the main active ingredient of turmeric. Studies suggest that curcumin may be an anti-fibrotic agents. However, curcumin on abnormal biological characteristics of hypertrophic scars and keloid fibroblasts with or without inhibition, the mechanism of how, whether for the treatment of hypertrophic scars and keloids, has not been reported. 【OBJECTIVE】 the curcumin concentration change of hypertrophic scars vitro into fibroblast proliferation and collagen synthesis, and explore its role in the treatment of hypertrophic scars mechanism. 2. Observation given different concentrations of curcumin treatment on cell proliferation and collagen synthesis of keloid fibroblasts in vitro, and preliminary analysis of the possible mechanisms of action. [Research]. Curcumin on cultured hypertrophic scar fibroblasts biological effects; 2. Curcumin on cultured keloid fibroblasts biological effects. Findings 1. Curcumin significantly inhibited the proliferation of fibroblasts in vitro hypertrophic scars and hypertrophic scars can induce apoptosis of fibroblasts; same time can inhibit apoptosis inhibitory gene Bcl-2 expression; curcumin treatment can significantly inhibit hypertrophic scar fibroblasts Ⅰ, Ⅲ procollagen mRNA expression; curcumin could significantly inhibit the proliferation of fibroblasts keloid into effective inhibition of collagen synthesis; curcumin could significantly inhibit the the keloid fibroblasts Ⅰ, Ⅲ procollagen mRNA expression, while inhibiting expression of connective tissue growth factor. 【Conclusion】 1. Curcumin vitro anti-proliferative scar, and has a role in promoting apoptosis, induced apoptosis mechanism may be related to the inhibition of apoptosis inhibition of Bcl-2 related to curcumin; 2. vitro anti keloid, its mechanism of action may be related to curcumin inhibition of connective tissue growth factor in the expression of keloid fibroblasts.
