Objective: To study the Toll-like receptor 9 (TLR9) expression levels in mononuclear cells (PBMCs) and serum IFN-α and IL-10 expression levels in the peripheral blood of patients with systemic lupus erythematosus analysis TLR9, IFN-α, IL expression level of correlation between -10 levels and SLE disease activity index (SLEDAI) correlation and IFN-α and IL-10, to explore their role in the the SLE pathogenesis of the clinical SLE new ideas. : From 60 SLE patients and 40 normal control group isolated PBMCs, using real-time quantitative PCR (real-time quantitative polymerase chain reaction, Real-time Q-PCR) detection PBMCs of SLE patients and normal control group in the Toll-like the receptor 9mRNA the expression levels; enzyme-linked immunosorbent assay (ELISA) detection SLE patients and normal control group, serum IFN-α and IL-10 expression levels; Pearson correlation or Spearman rank correlation analysis TLR9, IFN-α and IL -10 expression level of correlation between SLE disease activity index (SLEDAI) and IFN-α and IL-10, respectively. Results: Real-time Q-PCR test results show: both TLR9mRNA expression, the expression of SLE patients with active groups TLR9mRNA remission group and normal control group was significantly higher than in the SLE group and the normal control group, PBMCs difference was statistically significant (P lt; 0.01), and ease the differences between the group and the normal control group had no statistical significance (P gt; 0.05). ELISA test results show that: the SLE activity group IFN-α and IL-10 expression levels were significantly higher than the remission group, the difference was statistically significant (P lt; 0.01)), and the two groups are significantly higher than the control group, The difference was statistically significant (P lt; 0.01)). 3. Correlation analysis show: TLR9mRNA, the IFN-α and IL-10 expression water average with SLEDAI was being related (P lt; 0.01)); the IFN-α and IL-10 in the expression level was positive related (P lt; 0.01). Conclusion: 1.SLE active group of patients TLR9mRNA expression was significantly higher than the remission group and normal control group, the difference was statistically significant, remission was no significant difference between the group and the normal control group, suggesting TLR9 in the pathogenesis of SLE a certain sense. 2.SLE IFN-α and IL-10 expression level is higher than the normal control group, the difference was statistically significant, suggesting that patients with SLE regulatory T cells (Tr) enhanced the secretion of IFN-a and IL-10. 3. TLR9mRNA, of IFN-α and IL-10 the expression water average with SLEDAI was being related, prompt clinical can TLR9mRNA, IFN-α and IL-10 levels monitoring SLE disease activity in the case, as the SLE disease activity and prognostic indicators. IFN-α and IL-10 expression levels were positively correlated, the interaction between the prompt cytokines, affect each other.
