Experimental Study of Albendazole Nanoparticles Via the Portal Vein Administration in the Treatment of Echinococcus Multilocularis in Liver

Abstract


Objective:To compare the efficacy of albendazole (ABZ) nanoparticles in the treatment of hepatic alveolar echinococcosis (HAE) via the portal vein Infusion, oral and intravenous administration in Wistar rats. Method:80 successful HAE models made by ultrasound guided percutaneous and laparotomic route to inject echinococcus multiloculars suspension into the liver of Wistar rats. Rats were randomly divided into portal vein Infusion, oral, intravenous administration and the control groups. There were 20 rats in each group.12 hours after treatment by ABZ nanoparticles liver samples were gathered. Pathological examination and high performance liquid chromatography (HPLC) were performed. Results:(1)The positive rate and successful catheterization rate of ultrasound-guided percutaneous inoculation group were 60.00% and 93.75% respectively, that were higher than those of laparotomic inoculation group(75.76% and 42.86% respectively). The differences were statistically significant (P<0.05). (2) Degeneration was the main change in the oral and intravenous groups; the main pathological characteristics of the portal vein Infusion group was necrosis; the most of the HAE lesions were not changed after the procedure in the control group. Differences were significant between the portal vein Infusion and oral, intravenous groups (P<0.017).No statistically significant difference was detected between oral and intravenous groups(P>0.017).(3)The concentration of ABZ and ABZSX were the highest in the portal vein Infusion group. The differences were statistically significant from the portal vein Infusion group to oral and intravenous groups(P<0.017). No statistically significances was found between the oral and the intravenous groups(P>0.017). Conclusion:(1)Ultrasound-guided percutaneous inoculation technique is a simple method with high positive rate for HAE modeling. It can significantly decrease the mortality rate and suffering of animal. Can be used as the first line modeling way of the HAE experimental research.(2)ABZ nanoparticles via the portal vein Infusion is more effective than the oral and the intravenous to treat E.M in liver. The present study established a basic experimental data for interventional therapy through portal vein and hepatic artery.
