Abstract


OBJECTIVE: Use modern methods and technology research to analyze the effective elements and its content in setting relevant quality standards of kudzu vine extract on the basis of clearing the chemical constituents of kudzu vine extract further research on the role played of the prevention and treatment for osteoporosis by pharmacology.METHODS : Use thin-layer chromatography (TLC) and high-performance liquid chromatography(HPLC ) method to determine the component and content of kudzu vine extract .Choose qualified rats to test the protective effect of kudzu vine extract on osteoporosis with ovariectomized rat models and rat models induced by dexamethasone .Wistar rats were randomly divided into groups of the model group ,Xianlinggubao capsule, positive control group , kudzu vine extract high, medium and low dose group according to the weight, after administered 6 months the determination of mechanical properties of femur bone, calcium and phosphorus, serum estrogen level were measured.RESULTS: After six months of continuous administration, the rats were observed by the indexes, suggesting that Pueraria lobata extract extract significantly improved the general situation of the modeling rat, rats increased body weight, increased uterine and ovarian factor. Three treated groups were significantly improved resistance to external force of bone tissue,significantly improve the performance of the femur against external forces. Collected one side femur of the rats got dry weight and ash weight, calculated the ratio of wet weight, dry weight, ash weight and volume, suggesting that different treatment group was in different improved the osteoporosis performance caused by modeling. The three dose groups statistically had significant difference (P< 0.01). Pueraria lobata extract on serum calcium and phosphorus content was not obvious,the model group compared with the control group had no significant difference Calcium and phosphorus content, suggesting that serum calcium and phosphorus in the modeling had no effect. But kudzu vine extract can significantly decrease serum alkaline phosphatase in rats. Experiment also confirmed that kudzu vine extract can different increase the concentration levels of estradiol of rats. Significant increase in bone calcium and phosphorus. Also found that the chloride ion is also on the apparent intervention. Pathology suggested kudzu vine extract in some extent improved the changes in bone and pathological,increased trabecular number.CONCLUSION: The multi-angle study of different treatment levels of on osteoporosisrat model induced by dexamethasone,results suggest that kudzu vine extract has significant improvement.
