Purpose : The high-fat diet to establish non- alcoholic fatty liver rat model , the dynamic expression of L-FABP and PPAR-α in the detection of the modeling process at different time for clinical diagnosis , treatment and prognosis provided an important theoretical basis . Methods: Wistar male rats 84 , body weight (180 ± 10g), were randomly divided into a control group and model group . The control group diet basal diet, the model group eating high fat diet , the ratio is as follows : 87.8% of the basal diet , 10% lard , cholesterol 2% bile salts 0.2% . Each group was randomly divided into 0,2,4,8,12,16,18 weeks seven -phase group , in which the model group after 12 weeks diet basic feed it fatty liver in natural recovery . 7 , not phase blood from the heart to the determination of serum markers ( ALT , TG , CHOL, HDL -C , LDL- C was measured using automatic biochemical analyzer ) collected liver samples , part of the production of paraffin sections , HE staining . histopathological examination , observation of the degree of liver steatosis ; another part of the liver tissue using fluorescence quantitative RT-PCR method to detect L-FABP and of PPAR-αmRNA the dynamic changes , Western-blot detection of changes in the L-FABP . Results: control group rat liver L-FABP and PPAR-α in the expression of the white no statistically significant change . L-FABP mRNA in the first four weeks was significantly higher (P lt; 0.05), 8 and 12 weeks a very significantly higher ( P lt ; 0.01) , foundation diet for 6 weeks after the first 18 weeks was significantly decreased ( P lt ; 0.01) , but the contrast control group was significantly higher (P lt; 0.05). L-FABP protein 2,4,8,12 weeks were significantly higher (P lt; 0.01). Significant decline ( P lt; 0.05 ) the PPAR-αmRNA 4 weeks , 8 and 12 weeks was significantly decreased ( P lt ; 0.01) , foundation diet for 6 weeks after the first 18 weeks was significantly higher ( P lt ; 0.01) but the contrast control group was significantly decreased (P lt; 0.05). The first four weeks of the high-fat diet steatosis maximum area of 12 weeks , but no significant inflammatory response . The basal diet steatosis was significantly improved after six weeks . Conclusion: The high-fat diet fatty liver rat models were successful, and 12 weeks vesicular steatosis . L-FABP expression threshold value appears , and PPAR-α downregulation might play an important role in the process of the formation of fatty degeneration . Simple steatosis to some extent by dietary adjustments natural recovery .
