Abstract


Objective: Hepatocellular carcinoma (hepatocellular carcinoma, HCC) is one of the most common malignancy in the world . Tumor incidence and cancer mortality in our country , the survival rate , recurrence rate , early detection and early diagnosis key to treatment . Laser capture microdissection (laser capture microdissection, LCM) and gene chip two technologies emerging in the post-genomic era . The former capture a single cell or group of cells in the microscope under direct vision , successfully avoid tissue heterogeneity interference detection ; latter directly and quickly detect minute quantities of RNA can simultaneously monitor thousands of genes differentially expressed gene screening provide a powerful tool . In this study, coupled with the above two techniques to construct the gene expression profile of purified HCC tissue and normal liver tissue , screening HCC incidence related genes . Methods : 4 cases of single , non- metastatic HCC patient specimens first the LCM to obtain pure liver cancer cells and normal liver cells , and were extracted from traces of RNA, and identification of trace RNA quality using the Agilent 2100 Bioanalyzer . Quality inspection of micro- RNA linear amplification and cy3 , or cy5 two - color fluorescent labeling after , and then with the Agilent human 4 * 44K gene chip hybridization . Chips after washing , scanning the cy3 and cy5 Feature Extraction software Normalize standardized processing , and then filter out the candidate differentially expressed genes and HCC incidence . Statistics gene upregulation and down in all samples , filter out genes turn sorting and classification, to investigate the expression of a significant increase and decrease in the first ten candidate differentially expressed genes with HCC relations . Results: In the four pairs of samples of gene expression profiles in common differentially expressed genes of the purified liver cancer and normal liver tissue 1361 607 upregulated genes and 754 downregulated genes . The top 10 was significantly up-and down differences in expression of genes ( 20 ) , the control literature known differentially expressed genes for HCC , 10 expression changes are consistent with other tumor research reported four in six HCC differentially expressed genes and / or other tumors were not reported . Conclusion: 1 , combined with laser capture microdissection with gene chip technology was successfully constructed HCC differentially expressed genes spectrum . Study of differentially expressed genes is expected to provide new diagnostic and therapeutic targets for HCC .
