The Physiological Effects of Opioids and Nature Polyphenolic Compounds on Glutamate-induced PC12 Cell Injury and Its Possible Mechanism

Abstract


Opioids are well-known analgesics.Endomorphin1(endomorphin1,EM1), endomorphin2(endomorphin2,EM2) and morphine(Morphine,Mor) have high affinity and selectivity on theμ-opioid receptor and their unique physical activity has become a hot biology.Similarly,nature polyphenolic compounds like caffeic acid phenethyl ester (CAPE),caffeic acid(CA),ferulic acid(FA) and ethyl ferulate(EF) can play an important role in inhibiting oxidative stress.Therefore,we used PC12 cell treated with glutamate to simulate the excitotoxicity injury model and attampted to study the impact of EM1/2 and Mor on the cell activity in injury.In addition,we respectively disscused the physiological effects of opioids and nature polyphenolic compounds on glutamate-induced PC12 cell injury and its possible mechanism by detecting cell activity and SOD、GPx、GR and GSH activity.The results indicated that:1.100μmol·L-1 endomorphin 1,endomorphin 2 and 10μmol·L-1 morphine had highly significant impact on the cell activity of PC12 cell treated with glutamate(P<0.01). Administration of 1 mmol·L-1 naloxone(Nal),a nonselective opioid receptor antagonist, could restrain the cell viability.The effect of EM1/2 and Mor can be restrained by Nal, which indicated that it was mediated by opioid receptor and may play an important role in the prevention and treatment of neurodegenerative diseases.2.1μmol·L-1 CAPE,CA,FA and EF had highly significant impact on the cell activity of PC12 cell treated with glutamate(P<0.01).When 1μmol·L-1 CAPE,CA,FA and EF were administrated to glutamate-treated cells,the cell viability remained the highest,and the potency was CAPE>CA>FA>EF.3.When 1μmol·L-1 CAPE,CA,FA and EF were administrated to glutamate-treated cells,SOD and GPx activity decreased but the activity of GR and GSH increased.The mechanism of the physical effect of CAPE,CA,FA and EF may be mainly related to their antioxidative activities,which may also play an important role in the prevention and treatment of neurodegenerative diseases.
