Abstract

Male Sterility many higher plants, the most important result of this phenomenon is one of the reasons the anthers and pollen dysplasia, anthers and pollen development to become an important area of ??research in plant developmental biology. Will lead to male sterility mutant the corresponding gene cloning out and study its function in the anthers and pollen development process is an important method to study the molecular mechanism of anther and pollen development. Male sterile mutant of Arabidopsis mutant ma from a T-DNA insertion mutant library was screened controlled by a single recessive gene. Cytological observation of resin sections mutant ma anther small spores develop abnormal, and ultimately unable to form normal pollen grains. We use map-based cloning method to locate the gene in Arabidopsis T-DNA mutant missing on the first chromosome 23283kb with 23360kb between, apart 76kb. In this range, there is a newly reported male fertility gene ACOS5, proof of our work, ma is the acos5 a bit of mutants. This work for the MA cloning and study its structure and function to lay the foundation. The MtN3/saliva gene family of integral membrane proteins containing two MtN3/saliva transmembrane domain, and its members are widely exist in eukaryotes. At present, the family function less unclear molecular function of its transmembrane domain. 18 family members in Arabidopsis, in addition to the recently cloned RPG1 other MtN3/saliva gene function are unclear. A more comprehensive analysis of this study the Arabidopsis MtN3/saliva gene family, including gene structure, chromosome distribution, protein domains, phylogenetic relationships, gene expression profiling. Quantitative real-time PCR analysis of the four genes predicted highly expressed in anthers. The main results are as follows: 1. Comprehensive analysis the TAIR and a Pfam database in the gene nucleotide and amino acid sequence of the 18 Arabidopsis MtN3/saliva family the TAIR (http://www.arabidopsis.org) database. TAIR chromosomes drawing tools (Chromosome Map Tool) MtN3/saliva family gene mapping to the five chromosomes. 2. Use of the genetic structure of Gene Structure Display Server (http://gsds.cbi.pku.edu.cn/index.php) will the TAIR get 18 Arabidopsis MtN3/saliva family gene plotted in Fig. 3. Use of TMHMM Server v 2.0 Home (http://www.cbs.dtu.dk/services/TMHMM/) parameter is set to the default, predict protein structure diagram out the Arabidopsis MtN3/saliva family genes. 4 using the neighbor-joining method (neighbor-joining), Mega software (version 4.1) to construct a phylogenetic tree of the Arabidopsis protein sequence composed of repeated sampling from 1000 bootstrap values ??(bootstrap). 5 e-FP Browser the link provided by TAIR data is consolidated Genevestigater on expression profiling pairs the predicted MtN3/saliva family analyzed finishing, the main use of the data provided by the e-FP Browser and found that 4 (RPG1 except ) flowers highly expressed genes using quantitative RT-PCR technology of these four genes in Arabidopsis expression in each tissue analysis confirmed efficient expression of the three genes in flowers. These results provide a reference depth understanding gene family in MtN3/saliva.
