Abstract


PurposeThe rabbit limb ischemia-reperfusion injury (ischemia-reperfusion injure, I/RI) animal models were established with the method introduced by Feng Feng, etc [1] to observe the curative effect of Danshen on rabbits limb ischemia-reperfusion injury after Danshen intravenous injection in rabbit ear. To study the effects of Danshen on ischemia-reperfusion injury and to lay the foundation for further development of the Danshen used to prevent and cure limb ischemia-reperfusion injury.Method:48 Japanese white rabbits were used, weight 2.5±0.5kg, male or female, to establish rabbits limb ischemia-reperfusion injury model. The 48 rabbits I/RI models were divided into 3 groups randomly, namely, ischemia-reperfusion group (group A), the treatment intervention group (group B), and sham operation group (group C). Danshen intraperitoneal injections were given 30mins before modeling the treatment intervention group (group B). After anesthesia, we drew abdominal aortic blood, tested serum adenosine, SOD, MDA expression of Salvia intervention group and the ischemia reperfusion group after modeling, at time point 1h,2h,4h,24h.Result:1. Rabbit limb ischemia-reperfusion injury (ischemia-reperfusion injure, I/RI) animal models were established Successfully;2. MDA’s test results:SM treatment group (group B) and ischemia-reperfusion group (group A) were higher than those in sham operation group (group C), and the differences were significant statistically (P<0.01). MDA content in serum and skeletal muscle of Ischemia-reperfusion group (A group) at various time points were higher than those in the intervention group, the differences were significant statistically (P<0.05); and within 4h, gradually increased, at 24h, decreased.3. SOD test results:serum and skeletal muscle SOD activity of ischemia-reperfusion group (group A) and the treatment intervention group (group B) were lower than those in sham operation group (group C), the differences were significant statistically (P<0.01); and within 4h, gradually increased, at 24h, increased again. The intervention group (group B) adapted after drug intervention groups in the corresponding time point was higher than ischemia-reperfusion group (group A), and the differences were significant statistically (P<0.05).4. CPK, LDH test results:The serum CPK, LDH values in ischemia-reperfusion group (group A) and the treatment intervention (group B) were higher than those in sham operation group (group C), the differences were significant statistically (P <0.01), and their CPK, LDH values increased gradually with the reperfusion time prolonged within 24h. The intervention group (B group) in the corresponding time point was lower than ischemia-reperfusion group (group A), and the differences were significant statistically (P<0.05)5. Apoptotic index:ischemia-reperfusion group (group A) and the treatment intervention group (group B) had a different number of apoptotic cells, but the intervention group (group B) had less than the ischemia-reperfusion group (group A) in the corresponding time, and oxygen free radicals compared with the number of apoptotic cells (AI) and oxygen free radical levels were correlated significantly. Less apoptotic cells were found in Sham operation group (group C) and did not change over time.Conclusion:1. Danshen can reduce MDA content and promote the release of SOD after the rabbit skeletal muscle ischemia reperfusion injury2. Danshen can slow down or inhibit cell apoptosis in I/RI.3, Danshen has a significant protective effect on limb ischemia-reperfusion injury in that it might reduce the production of reactive oxygen species, increase antioxidantcapacity and alleviate lipid peroxidation, improve hemorheology and microcirculation status etc.4, The exact mechanism of action, time for intervention and dosage of Danshen on limb ischemia-reperfusion injury needs further study.
