Abstract


Cyanine dye has a long history , and has been widely used in optical materials . With the development of science and technology in recent years, people gradually applied in the biomedical field , and the emergence of a large literature associated . Cyanine dyes in nucleic acid determination , protein analysis, the stain as living cells , tumor detection and cancer cells to identify , as markers of biological macromolecules and photodynamic therapy , and describes the recent years, the emergence of new cyanine dyes The methods for their synthesis . Synthesized six kinds of nitrogen - containing heterocyclic intermediates , as raw materials and intermediates, these heterocyclic indole aldehyde synthesis five kinds A cyanine dye and composite merocyanine dyes . Use of 1H NMR, IR, UV-vis and MS structure of the dye . The dyes in different solvents UV - visible absorption and fluorescence spectra of the nature , results show that : the solvent has no effect on the peak shape of the absorption spectrum of the same dye absorbance is not . Dye in a protic solvent , the wavelength of maximum absorption with the solvent dielectric constant increases while the blue-shifted Stokes displacement decreases with the solvent dielectric constant ; in the aprotic solvent , the wavelength of maximum absorption with the solvent refractive index the increase of redshift , Stokes shift with increasing solvent refractive index decreases . UV - visible absorption and fluorescence spectra of seven kinds of dye and DNA under physiological conditions , the interaction of BSA , the results showed that : three kinds of water-soluble under physiological conditions dimethyl cyanine dyes with DNA , BSA after UV - visible and fluorescence spectra almost unchanged ; the two IA cyanine dye and two composite merocyanine dyes with DNA after UV - visible wavelength of maximum absorption redshifts 4-6nm, 7-80 times increase in the fluorescence intensity , might as fluorescent probes for the detection of DNA ; the two IA cyanine dyes and two composite merocyanine dyes with BSA , UV - visible absorption and fluorescence spectra changed little . The two kinds - A cyanine dye as a fluorescent dye in living cells staining results showed that one of leukemia cells staining better . Two kinds of composite merocyanine dyes photodynamic experiment , two kinds of dyes inhibit the growth of K562 ( chronic myelogenous leukemia cells) , may be used in photodynamic therapy .
