Abstract


Screened out with strong antagonism of the rice blast fungus and several common plant pathogenic fungi actinomycetes strains isolated from different soil samples in the strain identification and optimization of fermentation conditions , substances isolated aspects of research , to used to biological control of plant diseases . (1) the use of diluted coating method, soil samples taken from different regions of 48 parts were separated to obtain actinomycetes 433 . Adopted the the plate confrontation Law screening , performed better antagonism of 13 strains of pathogenic fungi strains , accounting for 3% of the total separation . Combined inhibitory effect of fermentation filtrate of pathogens , screened strains of rice blast fungus strains with strong antagonism WM2-4, that the strain in vivo inhibition of rice blast fungus bandwidth degrees 22.0 mm ; fermentation filtrate of the Magnaporthe grisea inhibition zone diameter of 32.5 mm, the inhibition rate was 89.4% . ( 2 ) by the WM2-4 strains morphological characteristics , cultural characteristics , physiological and biochemical characteristics , of strains 16S rDNA sequence determination and phylogenetic analysis , the WM2-4 was identified as the bright yellow Streptomyces ( Streptomyces galbus ) . ( 3 ) The strain the WM2-4 has a wider antibacterial spectrum , for a variety of plant pathogenic fungi such as Botrytis cinerea , Rhizoctonia , Sclerotinia , eggplant tenuis , Fusarium graminearum chain Fusarium sp , eggplant rot , rice blast fungus , Fusarium oxysporum , soybean Rhizoctonia root rot fungus of wheat and oats the sickle Jundeng have a certain extent. ( 4 ) fermentation condition optimization results show that the strain WM2-4 in the initial pH value of 7.5 , the incubation time of 5 d, inoculum size 6% 100 mL/500 mL medium volume , filtrate rice rice rice blast fungus The best inhibitory effect , inhibition rate of 69.2% . (5) acetone - cyclohexane (1:7, V / V) as eluent , the separation of the fermentation product of the strain WM2-4 column chromatography , to obtain an active material . The substance concentration of 1 mg / mL , the rice blast fungus inhibition zone diameter of 33.0 mm.
