Abstract


This study aims to explore the serum of patients with knee osteoarthritis (KOA) MMP-9 expression and significance, and to provide a basis for further study of the pathogenesis of osteoarthritis. Experimental collection Second Hospital of Jilin University, from June 2008 to November 2009 outpatient and inpatient osteoarthritis of the knee in patients with 30 cases for the experimental group, the other to take the medical examination of the Second Hospital of Jilin University health human serum 30 cases of normal control group. Using the ELISA method detected 30 cases of of KOA patients and 30 normal controls serum MMP-9, immediately after termination of the reaction at a wavelength at 450nm using a microplate reader scans the various specimens of MMP-9 concentration, according to the standard concentration and OD worth. SPSS13.0 software for statistical analysis and the ELISA experiment results (ng / ml) are expressed as mean ± standard deviation (x ± s), measurement data using t-test, P lt; 0.05 as statistically significant. Determination of serum MMP-9 expression levels of 30 cases of of KOA patients and 30 healthy people, the results were: 14.91 ± 4.909ng/ml 9.598 ± 2.396ng/ml; KOA early, mid-and advanced serum MMP-9 The average concentration: the 17.613 ± 5.186ng/ml, the 14.632 ± 4.599ng/ml, 11.915 ± 3.116ng/ml; the normal group male serum MMP-9 average concentration of 9.965 ± 2.42ng/ml; female serum MMP -9 average concentration of 9.23 ± 2.398ng/ml. The experimental group, the average concentration of male serum MMP-9 13.528 ± 4.469ng/ml; the the female serum MMP-9 average concentration for 16.119 ± 5.093ng/ml. The experimental group, under 50 years old (50 years old) the average concentration of serum MMP-9 for 15.261 ± 4.228ng/ml; over the age of 50 serum MMP-9 average concentration for 14.734 ± 5.311ng/ml. The normal control group, 50 years of age (50 years old) serum MMP-9 average concentration 9.55 ± 2.165ng/ml; and over the age of 50 serum MMP-9 average concentration of 9.66 ± 2.76ng/ml. The application of statistical analysis, the KOA serum MMP-9 levels were significantly higher than in the normal control group, showed that MMP-9 to participate and play a role in the KOA pathogenesis. KOA early serum MMP-9 level was significantly higher than the late, showed that MMP-9 in the KOA early, mid-play a role in degradation of extracellular matrix; Men and women in the experimental group between serum MMP-9 levels were relatively no significant difference not statistically significant, visible gender factors for serum MMP-9 concentration. The age factor in the experimental group, less than 50 years old (50 years old) compares with more than 50 years old serum MMP-9 levels, and no significant difference was not statistically significant, visible in the experimental serum MMP-9 little effect. Determination of MMP-9 level in the serum of patients with OA, lay the foundation for further study of the role of MMPs in the pathogenesis of osteoarthritis; help us in the early OA patients appear to make timely and correct diagnosis before clinical symptoms and radiographic change thereby providing more opportunity for the prevention and treatment of diseases.
