Abstract

Selection of healthy and disease-free, 4-month-old of similar weight F2 generation hybrid ewes (Dorset ♂ × Small Tail Han Sheep ♀) 36, were randomly divided into group A (low energy group), B group (energy group) group (high-energy group) Group 3, n = 12. Group B for the control group, fed diets designed in accordance with the energy in the sheep breeding standards of the U.S. NRC (1985) requirements, metabolic energy MJ 10.32 / d, group A and group C treatment is reduced on the basis of medium energy level and increased by 30%: i.e. a low energy level (A group) metabolizable energy 7.21 MJ / d, (C group) metabolism of the high energy levels can 13.48MJ / d, each group of proteins and other nutritional index substantially consistent. Fed 50 days from each experimental group were randomly selected six sheep to be slaughtered, measured before slaughter live weight, carcass weight, dressing percentage, backfat thickness, slaughter to take both sides of longissimus muscle (12 ribs) The hind legs biceps femoris measured intramuscular fat content. Take longissimus muscle immediately after slaughter, biceps femoris, heart and subcutaneous adipose tissue samples for extraction of total RNA, using real-time PCR method by optimizing the reaction conditions and the system of hormone-sensitive lipase (HSL) gene in longissimus muscle, the biceps femoris heart and subcutaneous adipose tissue in the expression level of detection. Three energy level of the diet of the sheep HSLmRNA absolute expression amount. The test results are as follows: a low energy group test sheep before slaughter live weight, carcass weight, dressing percentage was significantly lower than in energy and high-energy group (P lt; 0.01 or P lt; 0.05), in the energy group and the high-energy group The the slaughter performance difference was not significant (P gt; 0.05); backfat thickness of the low energy group test sheep was significantly lower than the energy, high-energy group (P lt; 0.01), in the energy group and the high-energy group differences between extremely significantly (P lt; 0.01). Low-energy group test sheep back longest muscle and biceps femoris intramuscular fat content in a very significantly lower than the energy, high-energy group (P lt; 0.01), the difference between in the energy group and the high-energy group is also highly significant (P lt; 0.01). 2 dietary energy levels affect sheep subcutaneous adipose tissue, longissimus muscle, the biceps femoris heart HSL gene expression, HSLmRNA expression is reduced with increasing dietary energy levels; three groups of energy in the subcutaneous fat the level between the HSLmRNA of absolute expression of a significant difference (P lt; 0.01), low energy group was significantly higher than in the high-energy group expression; expression of low-energy group in longissimus muscle, biceps femoris high-energy group, the difference is extremely significant (P lt; 0.01), a significant difference (P lt; 0.05) in energy between the high-energy group; difference between the energy group and the low, the high-energy group in the expression of the heart significantly (P lt; 0.05), low energy group was significantly higher than that of the high-energy group (P lt; 0.01). 3 HSL gene in sheep adipose tissue, longissimus muscle, with tissue-specific expression of the biceps femoris and heart, was the lowest in the highest amount of adipose tissue expression, heart, adipose tissue expression levels are extremely significant higher than back longest The expression of the muscle biceps femoris and heart expression (P lt; 0.01), another in the muscle tissue in the back dorsi muscle than the biceps femoris.
