Liver cancer is the most common malignant tumor, liver cancer early diagnosis is the key to improve patient survival. The current diagnosis of liver cancer mainly by including AFP (alpha fetoprotein, AFP) levels serological detection and combined liver imaging for detection of ultrasound contrast, but due to the lack of adequate diagnostic sensitivity and difficult to achieve early diagnosis of liver cancer. Therefore, the discovery of new serum protein biomarkers for clinical diagnosis of liver cancer has important value, the current through the analysis of HCC tissue and healthy tissue differences in protein expression levels, has found many potential protein biomarkers, but most have not been serum Identification of learning. Therefore, this article intends to carry out through carcinoma antibody microarray technology to the identification of potential serum markers studied. Objective: To establish a quantitative antibody-based detection technology through the chip, used in liver cancer potential identification of serum markers for early diagnosis and prognosis of HCC surveillance basis. Methods: Preparation of aldehyde-modified glass surface potential against 14 kinds of liver cancer markers in serum protein antibody microarrays, according to standard clinical procedures for collection of serum samples collected 44 cases of liver cancer and 46 cases of healthy serum samples, using Serum samples to be directly biotinylated antibody microarray analysis strategy, through the liver cancer in serum samples from healthy these potential protein markers of the expression level difference, to achieve these potential serological markers. Results: establish a directly labeled serum samples for antibody microarray analysis techniques optimal conditions: such as the use of 5% skim milk powder as a blocking agent of the chip, temperature blocked for 1 h; biotin labeled reagent and serum protein mass ratio of 1 : 40, serum samples 1:200 final dilution obtained when the ideal microarray results. Antibody Chip serum samples for 90 cases of the preliminary test results showed that HSP70, CK19, GPC3, PTMA, AFU, GP73, HBXAg, CA125 and other 8 kinds of potential protein marker expression levels in liver and serum of healthy subjects was significantly different, 8 kind of potential biomarkers serum protein markers in hepatocellular carcinoma, the positive predictive rate can reach 88.6%, specificity of 97.8%. Conclusion: The serum samples based on the direct biotin-labeled antibody microarray analysis strategy is an effective index can simultaneously detect multiple protein expression in serum through quantitative analysis techniques can be done not only 14 kinds of potential markers of liver serological identification, but can also be achieved by extending the research potential for more liver cancer markers as well as other diseases identified potential serological markers provide reference research tool.
