Objective: To investigate the expression of Ezrin protein and DNA ploidy analysis clinicopathological significance in cervical cancer. Materials and Methods: 15 cases of normal cervical epithelium, 51 cases of cervical intraepithelial neoplasia (cervical intraepithelial neoplasia, CIN), cervical glandular epithelial neoplasia (Cervical Glandular intraepithelial Neoplasia, CGIN), 53 cervical squamous cell carcinoma ( squamous cell carcinoma, SCC) and 5 cases of cervical adenocarcinoma (Adenocarcinoma) of 130 cases of cervical lesion specimens selected from the group consisting of Tongji University First Affiliated Fuyingyiyuan, Jilin Railway Hospital and Yanbian Fuyingyiyuan, Department of Pathology, 2000-2009 archived paraffin blocks cervical squamous cell carcinoma FIGO (Federation Internationale de Gynecologie et d'Obstetrique) installments: Ⅰ 11 cases, Ⅱ of 14 cases, 19 cases of Phase III, IV of nine cases; FIGO staging of cervical adenocarcinoma: Ⅰ two cases of cases, Ⅱ, Ⅲ of 2 cases. Cervical squamous cell carcinoma with lymph node metastasis in 28 cases, 25 cases without lymph node metastasis. Cervical cancer, (I - II a) of 24 patients with advanced cervical squamous cell carcinoma in 29 cases. Application the immunohistochemistry detection Ezrin protein in normal cervical epithelium, CIN, CGIN, cervical squamous cell carcinoma and cervical adenocarcinoma expression and application of flow cytometry cervical lesions DNA ploidy. Results: Ezrin protein expression was negative in normal cervical squamous epithelium and glands, a small part of the positive cases were mainly the Ezrin protein localization in normal cervical gland luminal surface of the inside of the cell membrane or cytoplasm in normal cervical epithelium positive was mainly located in the basal layer of the epithelium, cervical cancer and abnormal cells expressed as diffuse positive staining of the nucleus or cytoplasm. Ezrin protein expression in normal cervical epithelium was 13.3% (2/15), mainly as a weak positive, but in CIN the CGIN, cervical squamous cell carcinoma and adenocarcinoma of the positive expression rate was significantly higher positive rate 86.3% (44/51), 100% (6/6), 88.7% (47/53) and 100% (5/5). Ezrin protein positive expression rate in keratinizing and non-keratinizing cervical squamous cell carcinoma were 65.2% (15/23) and 80.0% (24/30) (P gt; 0.05); I - II A period (early ) and II B-IV (late) in cervical squamous cell carcinoma Ezrin strong positive expression rate of 70.8% (17/24) and 75.9% (22/29) (P gt; 0.05); lymph node metastasis and without lymph node metastasis of cervical squamous cell carcinoma of Ezrin strong positive expression rate was 96.4% (27/28) and 48.0% (12/25) (P lt; 0.05). In addition, DNA ploidy results showed that 15 cases of chronic cervicitis and 21 cases of CIN-I and CIN-II lesion specimens showed DNA diploid, in CIN III CGIN, invasive squamous cell carcinoma and invasive adenocarcinoma detected DNA aneuploidy, the detection rate was 18.8% (3/16), 16.7% (1/6), 49.1% (26/53) and 80.0% (4/5) with normal cervical epithelium and CIN-I and CIN-II lesions significantly different (P lt; 0.01). Conclusion: (1) Ezrin protein expression in cervical lesions can be positioned in a certain extent indicative of normal cervical epithelium to cervical intraepithelial neoplasia (CIN) and / or malignant transformation trend. The ② Ezrin protein detection can be used as indicators of early diagnosis of cervical cancer and precancerous lesions. (3) Ezrin protein overexpression important test for the determination of cervical cancer lymph node metastasis, combined with DNA analysis can be used as a prognosis of a detection index.
