Objective: To investigate whether estrogen impact injured area of ??brain tissue after traumatic brain injury in rats Bcl-2, Bax expression and brain edema brain, which play a protective effect. Methods: 75 healthy clean adult male Wistar rats were randomly divided into two groups. The first group of 60 rats for the detection of the injured area of ??the brain tissue of Bcl-2 and Bax protein expression and TUNEL-positive expression. The second group of 15 rats were used for the detection of brain water content. Each group was divided into a treatment group and a control group and trauma group. Treatment group: Feeney the freefall brain injury device injured animals, and given estradiol benzoate (1mg/kg · d) by intraperitoneal injection therapy treatment immediately after injury, after a day of treatment, until the animals were killed ; the control group: injured animals with the law, and intraperitoneal injection of normal saline treatment; trauma group: the same law injured animals not treated. Wait until the rats were sacrificed at each time point, take the injured area of ??brain tissue by immunohistochemical methods prosecution of Bcl-2 and Bax positive cells change with hematoxylin - eosin staining of brain tissue apoptotic morphological changes and calculated Bcl -2 and Bax positive cell ratio, application TUNEL staining of apoptotic cells was observed and calculated the apoptotic index and brain brain water content determination. Results: (1) HE staining light microscope: the structure of nerve cells in the treatment group was better than the control group and trauma group, no significant nerve nucleus pyknosis, nuclear fragmentation, nuclear chromatin edge set in, and arranged evenly distributed . (2) Light microscopy of Bcl-2 and Bax positive cells staining in the cytoplasm and (or) perinuclear few positive cells visible brownish yellow; mainly located in damage to adjacent areas, away from the damage zone; treatment group and trauma group, compared to the control group at each time point, Bcl-2 percentage of positive cells was significantly higher (P lt; 0.05), trauma and control groups was not significantly different (P gt; O.05); percentage of Bax positive cells of three groups compared with each other, No statistically significant difference (P gt; O.05); treatment group, Bcl-2 and Bax ratio higher than that in the control group and trauma group. (3) TUNEL staining positive expression with brown particles in the nucleus; treatment group, the apoptotic index was significantly less than the control group and trauma group (P lt; 0.05), trauma group and control group difference was not obvious (P gt; O. 05) (4) The three groups of brain water content by one-way ANOVA was significant difference (P lt; 0.05). Compared by pairwise between the groups, the treatment group and the control group, trauma group, the difference was statistically significant (P lt; 0.05), while the control group and trauma group showed no significant differences (P gt; O.05). Conclusion: (1) estrogen on the expression of Bcl-2 in the injured area of ??the rat brain after TBI promoted, while no significant effect on Bax expression. (2) estrogen can reduce the rat cerebral edema after TBI. (3) TBI estrogen by promoting the injured area of ??the rat brain expression of Bcl-2 and cerebral edema, which play a cerebral protective effect.
