Abstract


[Objective] a large number of studies have reported artery bleeding or intravenous injection of sodium nitroprusside (Sodium nitroprusside, SNP)-induced acute hypotension in anesthetized animals, possibly through the peripheral vestibular organ by the the vestibular nerve release of glutamate (Glutamate, Glu) excited vestibular medial nucleus (medial vestibular nucleus, MVN) and MVN, c-Fos protein expression. In awake animals is also able to appear the same results, in particular, is very little research on acute hypotension excited MVN Glu-NMDA receptor mechanism. This study was awake animals lead to the rat MVN neuronal excitability process NMDA receptor agonists (NMDA) blockers (MK-801) MVN within the c-Fos protein expression induced acute hypotension. [Method] weight 250 ± 50g male Wistar rats of clean grade 60. The animals were randomly divided into three large groups, the normal group of the vestibular organ; unilateral vestibular organ damage group; NMDA receptor group. First, the use of immunohistochemical methods to observe the normal animals and drugs destroy the side of the peripheral vestibular organ chronic phase (2w) (Aminobenzenearsonic salt), intravenous SNP (20μg/kg/min, 3min) induced acute low MVN, the expression of c-Fos protein in the blood pressure (50%), followed by combined pharmacological methods to further explore the NMDA receptor agonist (NMDA) injected into the lateral ventricle and blocker (MK-801) on the effects of to explore drug-induced the acute hypotension excited MVN glutamate NMDA receptor mechanism. [Results] In normal animals, the use of intravenous SNP blood pressure dropped by an average of 50% after 30min, 60min, 120min when the MVN District, c-Fos protein expression induced by acute hypotension and expression of c-Fos protein in 60min. the most obvious. Intravenous saline control group had no significant c-Fos protein expression compared to the differences between the two groups was significant (P lt; 0.001). 2 2w after unilateral vestibular organ damage induced acute hypotension 30min, 60min, 120min, no c-fos protein in the ipsilateral MVN District obvious expression. But the damage the contralateral MVN zone c-Fos protein still increased significantly compared with the corresponding time of the injury side showed a significant difference (P lt; 0.001), the most obvious expression of c-Fos protein in acute hypotension after 60min; And with vestibular organs of normal animals. 3, the vestibular organs of normal animals intracerebroventricular injection of NMDA blocker (MK-801, 1 mmol / L) induced acute hypotension after 60min, could not be seen in the MVN, a clear expression of the c-Fos protein and intracerebroventricular injection artificial cerebrospinal fluid group was not significantly different (P lt; 0.001). Vestibular organs of normal animals the intracerebroventricular injection NMDA agonist (NMDA, 1 mmol / L) 60min minutes later, bilateral the MVN District, c-Fos protein expression was significantly increased compared with artificial cerebrospinal fluid there is a significant difference (P lt; 0.001 ). [Conclusion] 1. Conscious animals intravenous SNP induced acute hypotension, can be excited MVN; destruction of the peripheral vestibular organ can block the the awake animals induced acute hypotension excited MVN role; acute hypotension induced in conscious animals the MVN excited effect in Glu-NMDA receptors are involved.
