Diabetes is a common chronic metabolic diseases, due to its high prevalence and mortality has gradually developed into a threat to human health following cancer, cardiovascular and cerebrovascular diseases after the third biggest killer. The research and development of both hypoglycemic drug can treat complications, diabetes drug development direction. China's use of Chinese herbal medicine to treat diabetes has a long history, but the mechanism is unclear, limiting its development promotion. This selection of Panax and Anemarrhena active ingredient as a model drug, the basis of previous work carried out as follows with the original innovative research: (1) the role of polar components in bovine serum albumin (BSA), the role of screening and protein components provide a theoretical basis, as further identify the active ingredients of Panax notoginseng. (2) the use of fluorescence spectroscopy to study the monomer saponin (Rb1, Rg1, Re) and its binary mixtures with BSA interaction mechanism between the two components are mixed in Chinese herbal medicine, exploratory research. (3) Comparative study of ultrafiltration, the method of equilibrium dialysis, gel filtration chromatography, etc., to study the role of drugs and protein binding, and to provide a reference for further study of the mechanisms of drug action, and tries to find the active ingredient in the Filter DRUGS simple and efficient method. The thesis is divided into five chapters. Chapter: a brief introduction to the overview of diabetes and herbal medicine in the treatment of diabetes, various methods of study drug binding of small molecules and proteins. Chapter II: rough points of the main active ingredient of notoginseng (Panax notoginseng saponins) using silica gel column chromatography, the role of polar components in bovine serum albumin (BSA), the use of methods such as fluorescence spectrometry screening and protein the role of the component, the application soft ionization mass spectrometry (ESI-MS) and infrared spectroscopy according to relevant literature values ??inferred main components and structure of each component may be. Chapter III: fluorescence spectroscopy in phosphate buffer solution (pH 7.4), respectively on a single component (ginsenoside Rb1 in Rg1, Re) and binary mixtures (Rb1 in-Rg1 and Rb1 in-Re, Rg1-Re) and the BSA interaction. The results of these experiments can provide the basis for the interpretation of the drugs used alone and mixed use pharmacological mechanisms at the molecular level, and to determine whether the combination of drugs with antagonism. Chapter 4: equilibrium dialysis, ultrafiltration combined with high performance liquid chromatography and gel filtration chromatography determination of mangiferin BSA binding rate for the first time, to compare different methods of measurement results prove gel permeation chromatography in the determination of protein - there are limitations on the rate of drug binding. Chapter: selection of polystyrene resin as a matrix resin to be modified, and bovine serum albumin grafted on the surface of the material, investigated the possibility of its adsorption capacity of the small molecules of the drug as well as a column packing.
