Abstract


Objective : a preliminary study of leptin in vitro mouse calvarial osteoblast proliferation and Collagen I (I collagen ) and Cbfal ( core binding factor 1) mRNA expression , leptin acts on osteoblasts may mechanism for the clinical use of drugs to provide a theoretical basis . Methods: a cultured mouse osteoblasts and observe its shape and function . 2 in mouse osteoblasts in vitro experimental model, using MTT ( MTT ) assay after 24 hours with different concentrations of leptin ( 0,40,80 and 160ng/ml) acting on the primary cultured rat into proliferation of bone cells , the OD value was measured . Three different concentrations of leptin ( 0,40,80 and 160ng/ml) intervention in primary cultured rat osteoblasts by RT-PCR was used to detect the expression of Collagen I and Cbfal level . 4 statistical methods of experimental data obtained are presented as mean ± standard deviation, using spss13.0 statistical software to analyze the experimental data , there is no difference in the groups overall single factor analysis of variance between groups were compared using LSD method analysis , p lt; 0.05 was considered statistically significant . Result: an in vitro cultured mouse osteoblasts mostly fusiform , alkaline phosphatase staining positive culture medium β- glycerophosphate and L- ascorbic acid calcium Akane red pigment staining. Two different concentrations of leptin ( 0,40,80 and 160ng/ml) acting on the primary cultured osteoblasts after 24 hours , can promote their added value , OD values ??were significantly increased (P lt; 0.01), the control group and the experimental group the experimental group between the two groups , the differences were statistically significant (P lt; 0.01). 3 through semi-quantitative RT-PCR, primary cultured osteoblasts Collagen I and CbfalmRNA expression . The results showed that leptin increases the expression of Collagen I and CbfalmRNA dose- effect relationship . Control group and the experimental group , the experimental group comparison between groups were statistically significant (P lt; 0.05). Conclusion: a leptin promote primary cultured osteoblasts . 2 leptin and Cbfal by adjusting the expression of Collagen I , thus promoting bone formation.
