Abstract


Embryonic stem cells (Embryonic stem cells) are a class of self-renewal in vitro specific culture conditions can be maintained , and having the ability to differentiate to the various cell types ( three germ layers ) totipotent cells . Traditional mouse embryonic stem cells cultured in vitro can not be separated from the leukemia inhibitory factor LIF ( Leukemia inhibitory factor ) , the LIF long-term scientific monopolized by foreign biotech companies and high price , has been plagued by scientists engaged in embryonic stem cell research . In order to solve this problem, in the pGEX-2T-MLIF transfected E. coli by IPTG induced synthesis of LIF and the separation and purification , then we have obtained with purified LIF is added to the mouse embryonic stem cells in a culture medium used for small mouse embryonic stem cells cultured in vitro . The experiments show that the in vitro culture of the late , separation and purification of LIF and commercialization of the LIF the same has to inhibit the differentiation of mouse embryonic stem cells and to maintain the characteristics of self-renewal . At the same time the mouse embryonic stem cells in nude teratogenic experiments further prove the LIF cultured embryonic stem cells can be used to maintain the pluripotency of embryonic stem cells .
