Abstract


Objective: canis is pro-animal fungi, often infected animals (especially dogs and cats) and humans, causing fungal disease. It is easy to scalp tissue infection, and is more common in children. In recent years, the close contact of humans and dogs and cats and other pets lead canis disease incidence is increasing. Canine spores pathogenic role in the process of the protease class with tinea capitis and tinea capitis pathogenesis is the research focus of attention. The experimental application of semi-quantitative RT-PCR technology to detect 30 canis tinea capitis strains and 30 tinea strains serine hydrolase (FSH1) expression levels; detection both in the sand castle liquid medium passaged FSH1 The expression and Children's scalp, before and after the induction of the smooth skin of children FSH1 expression. Expect that the results of this experiment for FSH1 function analysis to lay a foundation to provide a basis for FSH1 in the pathogenesis of tinea capitis caused by Microsporum canis. : 30 tinea capitis strains and 30 body ringworm strains inoculated in rice medium, cultured for 2 weeks separated spores. 5 × 106 spores inoculated in a liquid medium in 100 ml of sandcastles mycelium corporation culture in 21-28 days after the medium was collected, the first generation, fifth generation Five years after the pass using the same method. Tinea capitis the strains and tinea strains each 5 × 106 spores were inoculated in 100ml Children's scalp induction medium 100ml Children induction medium smooth skin, 21-28 days of culture mycelium collected the two skin tissue induction medium group. Semi-quantitative RT-PCR analysis the above strains FSH1mRNA expression level. Results: canis and in the sand castle liquid medium containing child scalp tissue, smooth skin and children two substrate induction medium growth, the formation of mycelial clumps. Bacteria growth in the sand castle liquid medium better growth slightly worse in the induction medium containing child scalp tissue and smooth skin tissues of children. Application of semi-quantitative RT-PCR, canis FSH1mRNA expression levels. Results found FSH1 canis has its expression, and tinea capitis strains and the body the ringworm strain FSH1mRNA expression level differences are significant (P lt; 0.01); no significant difference both in the sand castle after the passage of the liquid medium FSH1mRNA expression levels ( P gt; 0.05); obvious before and after the induction of skin tissue the strains FSH1mRNA expression level differences (P lt; 0.01), and children's scalp, the strains FSH1mRNA Children smooth skin induced expression levels significantly different (P lt; 0.01). Conclusion: 1.FSH1 canis active expression. 2 the tinea capitis the strains FSH1mRNA expression was significantly higher than the the tinea strain. By the liquid medium sand castle passaged strains can stably expressing FSH1, the passage without impact. Induced by the skin tissue, FSH1mRNA expression level was significantly higher, and the child scalp tissue strains induced the expression of the most obvious. 5.FSH1 may cause tinea capitis related canis.
