Abstract


Background: Gastric cancer morbidity and mortality worldwide today one of the most malignant, ranking first in the digestive tract malignant tumors in China. Treatment of gastric cancer for the past 30 years has made considerable progress, however, still hovering around 20 to 30% 5-year survival rate of gastric cancer after at home and abroad. Tumor metastasis, seriously affecting the survival of patients. This topic based on HPA tumor metastasis, orthotopic transplantation model explore sputum dissipated junction side of tumor metastasis, in-depth study Xiaotan Sanjie Fang anti metastasis mechanism. Objective: To establish nude mouse human gastric adenocarcinoma MKN-45 orthotopic transplantation tumor model, through the application Xiaotan Sanjie Recipe intervention research Xiaotan Sanjie Recipe rat model of peripheral blood the of CK-20mRNA and tumor tissue MVD and HPA explore the possible mechanism of Xiaotan Sanjie Prescription in inhibiting tumor metastasis. Method: 1. Orthotopic transplantation of human gastric cancer MKN-45 tumor strain in BALB / C nude mice gastric parietal serosa observed transplanted tumors in mice after the survival of the state. Completely random approach 30 nude mice model were divided into three groups, namely: saline group, phlegm Sanjie Fang group ,5-Fu group (n = 10). The animals were sacrificed, the abdominal cavity was opened, collected in situ tumor tissue, known as tumor weight of the inhibitory rate was calculated to calculate the number of liver metastases. 4. Immunohistochemical staining in the tumor tissues, calculate the tumor tissue MVD measurement and analysis of orthotopic transplantation tumor tissue HPA expression strength. Results: 1. Successfully established human gastric cancer MKN-45 orthotopic transplantation model. The second week after the date of the model, partially nude mice can hit in the upper abdomen hard lumps after mass increases, the hard texture of necrotic tissue in the middle of the greater. 100% success rate of transplanted tumors in situ. Determination of tumor weight inhibitory rate was calculated Influence of Xiaotansanjie recipe ,5-Fu group inhibition rates were 56.3%, 47.2%, the statistical analysis showed that, Phlegm Sanjie Recipe group ,5-Fu group and the saline group significantly reduce the quality of the transplanted tumor, the difference was statistically significant (P = 0.000, P lt; 0.05); while Xiaotan Sanjie Recipe group than in the saline group liver metastases have decreased significantly, and the difference was statistically significant (P = 0.007, P lt; 0.05) :5-Fu group and the saline group differences were statistically significant (P = 0.03, P lt; 0.05); while no statistical difference between the the Influence of Xiaotansanjie recipe group and 5-Fu group significance (P = 0.538, P gt; 0.05). 2 Xiaotan Sanjie Fang group compared with the saline group, phlegm Sanjie can only significantly reduce the peripheral blood CK20-mRNA expression (P = 0.004, P lt; 0.01), the difference was statistically significant; while 5 -Fu group was not significantly reduced (P = 0.567, P gt; 0.05). 3 The results showed that the group Xiaotan Sanjie Recipe can significantly reduce tumor MVD (P = 0.00, P lt; 0.05), the difference was statistically significant; 5-Fu group did not reduce the role (P = 0.584, P gt; 0.05). Influence of Xiaotansanjie recipe group and the saline group tumor tissue in HPA expression, phlegm Sanjie Fang group than in the saline group can significantly reduce the tumor tissue HPA expression was significantly reduced, the difference was statistically significant (P = 0.01, P lt; 0.05); while the role of 5-Fu group were not significantly lower (P = 0.248, P gt; 0.05). Conclusion: 1. Use of human gastric adenocarcinoma MKN-45 tumor strain successfully established orthotopic transplantation model, phlegm Sanjie Recipe on orthotopic transplantation tumor growth was significantly inhibited, while effective in reducing liver metastases number; 2. Xiaotan Sanjie to effectively reduce the peripheral blood CK20mRNA expression, significantly inhibited tumor cells into the peripheral blood, has a significant inhibition of tumor metastasis: 3. Xiaotan the of Sanjie order to effectively reduce the tumor-bearing mouse tumor HPA suppression in tumor tissue, tissue MVD; 4. Xiaotansanjie recipe on the growth of human gastric cancer in situ xenograft model.
