Objective: To explore ursodeoxycholic acid on rat liver regeneration and cyclin D1 mRNA and protein expression . Methods: Male Wistar rats were divided into a control group and the group of ursodeoxycholic acid (UDCA group ) , control group received standard diet , UDCA group was given 0.1% UDCA feed , 7d underwent 70% partial hepatectomy (PH). 0,1,2,3 d at four time points after surgery , rats were sacrificed by immunohistochemical assay of liver tissue proliferating cell nuclear antigen PCNA expression ; Western blot assay using cyclin D1 protein expression ; RT - polymerase chain reaction (RT-PCR) detection of liver tissue cyclin D1 mRNA expression . Results: The two groups were in 0d almost no expression of PCNA protein , after PH 1d, PCNA protein labeling index increased rapidly and reached the peak , while the UDCA group PCNA labeling index was significantly higher (40.70 ± 4.73 vs 33.24 ± 5.59, P lt; 0.01). 2 d time , UDCA group PCNA labeling index remained significantly higher (35.6 ± 5.86 vs 29.45 ± 7.04, P lt; 0.05). 3 d , the two groups had no significant difference (P gt; 0.05). UDCA group in PH patients 1,2 d cyclin D1 mRNA expression and protein expression was significantly higher (P lt; 0.05), 3 d no significant difference between the two groups . Conclusion: ursodeoxycholic acid can promote rat liver after partial hepatectomy cell proliferation and increased cyclin D1mRNA and protein expression .
