Abstract


Objective: The study of genistein on human pancreatic BXPC-3 cells in vitro growth inhibition and apoptosis-inducing action . Methods: Using a cell culture method to establish stable passaged pancreatic BXPC-3 cell lines . The proliferation of the MTT assay genistein BXPC-3 cells . Flow cytometry was used to the determination of genistein after the change of the cell cycle and apoptosis . RESULTS: MTT assay showed that different concentrations of genistein role 24h in BXPC-3 cells , 48h after the difference between the experimental and control groups were statistically significant ( P lt; 0.01 ) . High concentration inhibition rate of lower concentration group high inhibition rate at 48h compared with the role of the the 24h group 's high . Flow cytometry analysis showed that genistein can make changes in cell cycle G2 / M phase cells increased with increasing concentration , and with time , while the decrease in the proportion of S phase cells in G0/G1 phase , G2 / M phase The increase in S phase , G0/G1 phase to reduce more obvious . Genistein can promote cell apoptosis and the apoptosis rate increased with increasing concentration , and incubation time after 48h , the apoptosis rate increases , and the difference between groups was statistically significant (P lt; 0.05). Conclusions : Genistein can inhibit proliferation of pancreatic cancer BXPC-3 cells and induced apoptosis , both were tested time and dose-dependent .
