Abstract


Object:By observering effect of Yangganhexue formula on sex hormones and the expression of ovarian Bcl-2 and Bax in primary senile female rat, we discuss mechanism of action of Yangganhexue formula which could prevent and cure perimenopausal syndrome, and testify the important theory from the liver in premenopause, This research may offer a new evidence of theory and practice about preventing and treating premenopausal syndrome.Methods:10-11 months old Wister female rats whose estrus elongation were set as primary senile female rat model by observating the cell of vagina and randomized into blank group, model group,Yangganhexue formula group, Liuweidihuangtang as positive control group. Besides, took 10 Wistar female rats of 4 months to be the normal contrast group. After all the groups were intragastric administrated for fifteen days, get the blood sample from abdominal artery. then test the level of serum Estradiol (E2),Follicle stimulating hormone(FSH), Luteinizing hormone(LH)by radio-immunity method. Then immediately separation of both ovaries, the expression of Bcl-2 and Bax protein in left ovarian cells was detected by the technique of Western blot, calculate the ratio of Bcl-2/Bax. The right ovaries were buried by paraffin and treated by HE to observering their change of morphosis.Results:Compared with young control group, model group increased the level of FSH and LH and decreased the level of E2 in blood, and decreased the expression of Bcl-2 and increased the expression of Bax obviously in ovarian cells, and increase the ratio of Bcl-2/Bax, the difference was significant(P＜0.05). Compared with model group and Liuweidihuagtang group, Yangganhexue formula can effectively increased the level of E2 and decreased the level of FSH and LH in blood, and increased the expression of Bcl-2 and decreased the expression of Bax in ovarian cells(P＜0.05), which improved ovarian morphosis.Conclusions:The results showed that Yangganhexue formula could adjust sex hormone level in blood, and so as to delay the aging of HPOA; which could inhibit apoptosis in rat ovarian and improve the ovarian morphology, and so as to delay ovarian aging. At primary senile stage of female rat, that Yangganhexue formula was better than Liuweidihuangtang testified the important theory from the liver in premenopause.
