Abstract


Background:Water is one of the most common things on the earth, and also is the most indispensable nutrient. Water accounts for 60% in the normal body and is the biggest part of it. As the medium for the body’s traffic of nutrients and waste products, water is nearly a universal solvent. And it brings to each cell the exact ingredients the cell requires and carries away the end products of its life-sustaining reactions. In our body, water is not simply a river coursing through the arteries,capillaries and vein. Some of water is part of chemical structure of compounds that form the cells,tissues and organs of the body. Water also takes part in many chemical reactions.It’s well known that water is composed of the elements hydrogen and oxygen. Hydrogen has three isotopes, they are protium (H), deuterium (D) and tritium (T). Protium contains a proton without neutron, while deuteron contains a proton and neutron, so the quality of D is twice that of H.D/H is about 1/6600 and D is about 150ppm in the nature. A world wide investigation about hydrogen’s isotope in precipitation water shows that D is about 120 to 160ppm. Deuterium is stable isotope of hydrogen and also a trace element. D2O and H2O have completely opposite influence to our body.D2O can slow down the cancer cell’s proliferation and some experiments told us that it has obvious inhibitory effect to the cancer transplanted mice, the mechanism may be related to inducing the block of lung cancer cell’s S stage and its apoptosis. A investigation in Japan showed that supplementation with hydrogen-rich water may have a beneficial role in prevention of T2DM and insulin resisitance by providing protection against oxidative stress However, there is no report about D2O’s antioxidation. As the rapid development of society, our living standards have increasingly improved. The unremitting pursuit to longevity makes antiaging research become a hot theme in science field. Senescence is natural process. We have been puzzled about what caused the wrinkles, pains and deterioration of aging. In the search for causes, one basic theory has won over the most proponents, introduced by Denham Harman, MD, PhD, in the mid-1950s, holded that cells eventually broke down due to attacking by caustic molecules called free radicals which caused oxidative stress.This paper based on the latest research about the anti-oxidation, with the D-galactose to make the aging mice model, aimed to find the low-deuterium water whether have the activity of anti-oxidation, It also benefit to provide experimental basis for further study of the mechanism and to explore a kind of low-deuterium water healthy beverage.Objective:To study the antioxidant effect of the deuterium depleted water in different organs on D-galactose induced aging mice model.MethodsTwenty-four male ICR mice are equally divided into three group:normal control group, aging model group and experimental group. The aged model mice were made by injecting D-galactose continually. Mice in experimental group drank deuterium depleted water while other two groups drank tap water. After seven weeks, we tested some antioxidases and oxidant productions as follow:serum:activity of superoxide dismutase (SOD),content of malondialdehyde (MDA);myocardial tissue: SOD,MDA,monoamine oxidase (MAO), lactate dehydrogenase(LDH). brain:T-AOC, Na+-K+-ATPase, NO, LPO and MDA;liver:SOD, glutathione peroxidase (GSH-Px),MDA and MAO. Results1、In normal control group, the level of SOD,T-AOC, GSH-Px, Na+-K+-ATPase showed significantly higher than those in D-galactose induced aging group. The MDA,LPO contents and the activities of MAO,NO were lower compared with aging group(P<0.05).Animal aging model was obtained by injecting D-galactose.2、In experimental group, the level of SOD,GSH-Px,Na+-K+-ATPase showed significantly higher than those in D-galactose induced aging group. The MDA,LPO contents and the activities of MAO,NO were lower compared with aging group(P<0.05). The level of LDH in cardic musle did not have significant difference among these three groups. The activities of T-AOC in brain also have no significant differences between experimental group and aging group.Conclusion1、Animal aging model was established by injecting D-galactose. 2、The deuterium depleted water could improve the effect of antioxidation effectively in several organs.
