The first part of plasma essential amino acid concentration and the degree of autophagy correlation analysis Objective: To explore the plasma concentration of essential amino acids on the degree of autophagy. : Aged rats were randomly divided into group A (normal diet control group), B group (1 day free diet one day fasting), C group (5 days free diet for two days of fasting) three groups (n = 15 only). Reared three months after the in vivo state take 1mm3 liver tissue stored at 4 ℃; heart blood, anticoagulant, centrifuged, deproteinized supernatant -70 ℃ of preservation. TEM observations the number of autophagic vacuoles in liver cells; performance liquid chromatography (HPLC) detection of plasma essential amino acid content; correlation Spearman rank correlation analysis between. Results: the valine concentration autophagy extent correlation maximum (r = -0.779, P lt; 0.001); lysine minimum (r = -0.517, P lt; 0.01); phenylalanine methionine and leucine autophagy degree of correlation coefficients were -0.566, -0.527 and -0.531 (P lt; 0.01); remaining concentration of amino acids and autophagy degree of correlation. Conclusion: the extent of certain plasma concentration of essential amino acids and autophagy were negatively correlated, which valine, phenylalanine, methionine, leucine, and lysine concentrations greater impact white bite the degree of cell. The degree of autophagy by regulating the levels of plasma amino acids regulate autophagy in clinical applications in one direction. The second part autophagy role in the anti-aging effects of dietary restriction research purposes: to observe the aged rats liver autophagy levels and the survival time of rats under different dietary restrictions ways to analyze the relationship between the two. Method: grouping the same, rearing three months after the specimens, high performance liquid chromatography (HPLC) detection of plasma essential amino acid content; transmission electron microscopy to observe the number of autophagic vacuoles in liver cells; RT-PCR was used to detect MAP1-LC3 mRNA expression; the postoperative continue feeding rats, recording a time of survival, draw survival curves. Results: two restricted diet group compared with the control group, the five kinds of plasma essential amino acid concentration decreased significantly enhanced the autophagy degree of liver cells, significantly prolong the survival time of rats, the difference was statistically significant. Two dietary restrictions between the two groups showed no significant difference. The correlation analysis showed that the survival time and liver autophagy levels high positive correlation (r = 0.842, P lt; 0.01). Conclusion: by reducing the concentration of plasma amino acids to enhance the degree of autophagy may be an important mechanism for the anti-aging effects dietary restrictions, to provide experimental evidence for the guidance of mankind's new healthy diet.
