Tall fescue in AtNYE1, AtNAP homologous gene isolation and functional analysis

AtNYE1 Arabidopsis chlorophyll degradation process is an important regulatory genes . In this study, rapid amplification of cDNA ends (RACE) technique , in tall fescue (Festuca arundinacea Schreb.) Was isolated a AtNYE1 homologous genes FaNYE1, and functional analysis was carried out . FaNYE1 cDNA length of 1441bp, containing 834bp long open reading frame (open reading frame), encodes a protein with 278 amino acid residues of the polypeptide genome contains two introns and three exons. Sequence analysis showed that , FaNYE1 NYE1 barley in the highest similarity with the AtNYE1 a 62% similarity . The expression pattern for FaNYE1 analysis showed that aging and dark FaNYE1 could significantly induce the expression . FaNYE1 in Arabidopsis Functional analysis showed that , FaNYE1 able heterologous complementary Arabidopsis nyel-1 mutant phenotype stagnant green , and the complementary phenotype FaNYE1 expression is positively correlated constitutive overexpression could promote FaNYE1 the degradation of chlorophyll . The study suggests that , FaNYE1 encoding a similar AtNYE1 chlorophyll catabolism regulatory protein , in the regulation of tall fescue chlorophyll degradation process plays an important role . AtNAP is one kind of Arabidopsis transcription factor associated with aging , the aging process plays an important role in regulating . In this study, rapid amplification of cDNA ends , tall fescue was isolated in a AtNAP homologous genes FaNAP, and functional analysis was carried out . FaNAP length of 1305bp, 1023bp contains a long open reading frame encoding a 340 amino acid residues containing the polypeptide , which genome contains two introns and 3 exons. Sequence analysis showed that , FaNAP and from wheat, barley, rice, corn , crude goat grass , soybeans and other species NAP has high homology. The expression pattern for FaNAP analysis showed that aging and dark FaNAP could significantly induce the expression , but more strongly induced senescence . FaNAP in Arabidopsis Functional analysis showed that , FaNAP heterologous complementary to the mutant Arabidopsis nap stagnant green phenotype, constitutive overexpression FaNAP can significantly promote the degree of aging of plants and plant senescence and expression in vivo FaNAP positive correlation . The study suggests that , FaNAP encoding an Arabidopsis AtNAP similar aging -related transcription factor in the regulation of tall fescue leaf senescence should play an important role .
