Abstract


Objective To study the caveolin -1 ( CAV - 1 ) mRNA expression in normal and ultraviolet radiation cataract model rat lens to explore its role in the process of oxidation cataract . Methods 50 rats lens cultured in MEM culture medium 8h randomly divided into group A (control group 25 ) group B ( UV irradiation group 25 ) . Experimental group : ultraviolet irradiation lens 30min Add MEM culture medium , set at 37 ° C under 5 % CO 2 < / sub > incubator cultured in culture medium after 0, 6, 12, 24 and 48 , respectively, in h, the application of fluorescence realtime RT - PCR to detect lens caveolin -1 protein expression levels in the control group does not shine on the same way and under the same conditions to detect the expression of caveolin -1 . Results after 30 minutes of ultraviolet radiation , using RT-PCR, measured caveolin - 1 mRNA expression level decreased gradually with time . 0h : experimental group (0.5920 ± 0.1254) control group ( 1.0242 ± 0.1117 ) ( P < 0.05 ) compared the difference was significant ; 6H : experimental group (0.3914 ± 0.2070) control group (0.9662 ± 0.1286) (P <0.01 ) compared the two groups there was a significant difference in the experimental group (0.2658 ± 0.0938) ; 12h : control group ( 0.9248 ± 0.1545 ) ( P < 0.01 ) compared to the difference was significant ; 24h : experimental group (0.1244 ± 0.0953) control group ( 0.8840 ± 0.0710 ) ( P < 0.01 ) compared significant difference ; 48h : experimental group ( 0.0598 ± 0.0414 ) control group ( 0.8420 ± 0.1023 ) ( P < 0.01 ) compared to the difference was significant . Conclusion ultraviolet radiation impact rat lens epithelial cells and lead to cell caveolin -1 expression is reduced, the reduction of caveolin -1 have a role in oxidizing the occurrence of cataract . Speculated that caveolin transparent protective lens to maintain oxidative damage .
