Abstract


Objective: advanced gastric cancer NF-kappa B and Rb protein expression E-cadherin/catenin complex in metastatic lymph nodes and its clinical significance of cancer cells express, explore the NF-kappa B the Rb protein E-cadherin/catenin composite material the mechanism of gastric cancer metastasis and prognosis of patients. Sought at the molecular level, more effective methods of gastric cancer early diagnosis and treatment update. Method: using the the immunohistochemical peroxidase labeled strand mold avidin method (SP) were used to detect the NF-kappa B, Rb protein in the primary tumor the stove and E-cadherin/catenin complexes in 86 patients with advanced gastric cancer primary stove and its corresponding lymph nodes within the expression of the cancer cells, combined with the size of the tumor, invasion depth, degree of differentiation, histological type, lymph node metastasis and prognosis of patients with clinical and pathological factors such as a comprehensive analysis. Results: NF-kappaB, the Rb protein E-cadherin/catenin complex immunoreactive products are brown, NF-kappaB, Rb staining located in the cytoplasm or nucleus, E-cadherin, alpha-, beta-and gamma-catenin was localized in the cell membrane. NF-kappa B protein expression in 86 patients with advanced gastric cancer tissues positive rate of 81.40% (70/86) and Rb protein expression in 86 patients with advanced gastric cancer tissues positive rate of 65.12% (56/86), E-cadherin / catenin complex in the primary tumor only one case of normal expression, 85 patients with abnormal expression of abnormal expression rate of 98.84%, the positive rate was 63.95% (55/86) of the complex expression within cancer cells in the lymph node metastases. NF-B expression and the pathological type of the tumor cells (signet ring cell carcinoma, undifferentiated carcinoma, squamous cell carcinoma and adeno-squamous cell carcinoma due to small sample size, it is not statistics) are closely related, the positive expression rate in the tubular adenocarcinoma 85.96% (49/57), mucinous adenocarcinoma, the positive expression rate was 61.90% (13/21), the difference was significant (P lt; 0.05); closely related to NF-B expression and tumor cell differentiation, in high differentiation of gastric cancer in the positive rate was 91.43% (32/35), poorly differentiated gastric expression was 74.51% (38/51), the difference was significant (P lt; 0.05); NF-B expression and tumor size and tumor invasion depth, the positive expression rate was 83.08% (54/65) in the tumor diameter greater than 5cm in diameter less than 5cm from the tumor positive expression rate was 76.19% (16/21), the difference was not significant ( P gt; 0.05); tumor depth of invasion invading the serosa positive expression rate was 82.05% (64/78), tumor depth of invasion is not invading the serosa positive expression rate was 75.00% (6 / 8), the difference was not significant (P gt; 0.05) Rb protein expression and tumor cell differentiation is closely related to the positive expression in well-differentiated gastric cancer was 85.71% (30/35), poorly differentiated gastric expression was 50.98% (26/51), the difference significantly (P lt; 0.05); Rb protein expression and pathological type of the tumor cells (signet ring cell carcinoma, undifferentiated carcinoma, squamous cell carcinoma and adeno-squamous cell carcinoma, it has not been due to the small sample size statistics) has nothing to do in the tubular gland cancer positive expression rate was 70.18% (40/57), mucinous adenocarcinoma positive expression rate was 71.43% (15/21), the difference was not significant (P gt; 0.05); Rb protein expression with tumor size and dependent on the depth of tumor invasion, tumor diameter greater than 5cm positive expression rate was 60.00% (39/65), the diameter of less than 5 cm from the tumor positive expression rate was 80.95% (17/21), the difference was not significant (P gt ; 0.05); tumor depth of invasion invading the serosa positive expression rate was 74.36% (50/78), tumor depth of invasion is not invading the serosa positive expression rate of 75.00% (6/8) the difference was not significant (P gt; 0.05) E-cadherin/catenin complex re-expression of tumor cells in the metastatic lymph node cancer cells is closely related to the degree of differentiation, in well-differentiated gastric cancer expression was 77.14% (27/35), the expression in poorly differentiated gastric cancer the positive rate was 54.90% (28/51), the difference was significant (P lt; 0.05); E-cadherin/catenin complexes are re-expressed in the cancer cells of metastatic lymph nodes and the pathological type of the tumor cells (signet ring cell carcinoma, undifferentiated carcinoma, squamous cell carcinoma and adeno-squamous cell carcinoma due to small sample size, it is not statistics) independent expression rate was 73.68% (42/57) in the tubular adenocarcinoma, mucinous adenocarcinoma positive expression rate was 52.38% ( 11/21), the difference was not significant (P gt; 0.05); E-cadherin/catenin complexes are re-expressed in the cancer cells of metastatic lymph nodes, tumor size and tumor depth of invasion, tumor diameter greater than 5cm expression rate of 64.62% (42/65), in the depth of invasion invading the serosa tumor diameter less than 5 cm from the tumor positive expression rate was 61.90% (13/21), the difference was not significant (P gt; 0.05); The positive expression rate was 74.36% (50/78), tumor depth of invasion is not invading the serosa positive expression rate was 62.50% (5/8), the difference was not significant (P gt; 0.05) (see Table 2). Kaplan-Meier curves of NF-kappa B, Rb protein expression and the prognosis of patients independent (P gt; 0.05) (see Figure 1 and 2); the E-cadherin/catenin complex expression in metastatic lymph nodes cancer cells and the patient's prognosis (P lt; 0.05) (Figure 3). Wilcoxon's log-rank test for difference between groups test showed no significant difference among the groups (P GT; 0.05) (Table 3). Cox's proportional-hazard multivariate analysis show E-cadherin/catenin complex expression in metastatic lymph nodes cancer cells can be used as an independent prognostic factor (P lt; 0.05) (Table 4). Spearman rank correlation analysis showed that NF-kappa B, Rb protein expression E-cadherin/catenin complex expression in metastatic lymph nodes cancer there was no significant correlation (Table 5, 6, 7). Conclusion: NF-kappaB the Rb protein E-cadherin/catenin complex may be associated with the occurrence, development and metastasis of gastric cancer is closely related to the presence of NF-kappa B; gastric Rb protein expression of metastatic lymph node cancer cells in the presence of E-cadherin / catenin complex objects high expression; E-cadherin/catenin complex expression is closely related to the prognosis of patients can be used as an independent prognostic factor in metastatic lymph nodes cancer, NF-kappa B, Rb protein expression prognosis; NF-kappa B, Rb protein expression E-cadherin/catenin complex expression in metastatic lymph nodes cancer among no significant correlation. Clinical detection of NF-kappa B, Rb protein expression E-cadherin/catenin complex in metastatic lymph nodes within cancer cells express sensitivity to improve early diagnosis and early treatment and improve the prognosis.
