The Influences of Donor Bone Marrow Cells Injection upon Prolongation of Pancreatic Allograft Survival in Rats

Abstract


Objective To explore the donor bone marrow cells induced input on the role of pancreas transplantation tolerance . Methods Male SD rats and female BN rats were used as experimental for receptors, receptor BN rats with streptozotocin -induced diabetic rats into and randomly divided into four groups : Group 1: after transplantation and transplantation without any special treatment ; 2 groups: recipient mice on day 1 after transplantation by the cyclophosphamide 150mg / kg body weight by intraperitoneal injection ; 3 groups : the day of transplantation , portal vein injection of donor bone marrow cells 2. 0 × 108 month; 4 group: the day of transplantation , portal vein injection of donor bone marrow cells 2. 0 × 108, while the first day after transplantation by the cyclophosphamide 150mg / kg body weight by intraperitoneal injection . After 14 days of each group was detected by PCR in peripheral blood donor rat -derived Y chromosome -specific fragments , while spleen cells by flow cytometry of donor -derived cells . Rats in each group and compared graft survival time . Results 4 groups can be detected in peripheral blood of donor origin SRY gene in rat spleen cells were detected in the cells of donor origin , while the other three groups were not detected . Meanwhile, the four groups of graft survival time of pancreatic function (18 ± 2.2) d, significantly longer than one (P lt; 0. 01). 2 groups (8.2 ± 1.6) d and 3 groups (8.8 ± 1.4) d with a group, graft survival time of pancreatic function was no significant difference (P lt; 0. 01). Conclusion The short-term use of immunosuppressive agents after transplantation and transplantation of donor bone marrow cells enter the day , can prolong the survival time of pancreas transplantation in rats . This result is bone marrow cells induced chimerism and tolerance related .
