Abstract


Objective: To study tumor metastasis gene MTA-1, the cytoskeletal link protein Ezrin expression levels in breast cancer tissue and normal breast tissue and pathological type, clinical stage, lymph node metastasis, the relationship between clinicopathological parameters and their the correlation between the analysis of breast cancer metastasis, prognosis, and to explore the possible molecular mechanism of MTA-1 to promote breast cancer metastasis. Methods: Xiangya Hospital Breast Surgery from March 2001 to March 2003 surgery for breast cancer specimens of 80 cases of pathologically confirmed 15 cases of normal breast tissue is selected as a control. Were used in situ hybridization and immunohistochemical staining SP method MTA-1 mRNA in 80 cases of breast cancer and 15 cases of normal breast tissue, Ezrin protein expression levels, both with clinicopathological analysis type, patient age, clinical stage, lymph node metastasis clinicopathological parameters, and to analyze the correlation between breast cancer in the MTA-1 mRNA, Ezrin protein and prognosis of breast cancer. Positive rate comparison of the count data crosstable chi-square test, the MTA-1 mRNA, Ezrin expression of the Spearman correlation analysis, the MTA-1 mRNA, Ezrin expression of the situation on the disease-free survival using the Kaplan-Meier survival analysis and Log- rank test. Results: (1) normal breast tissue MTA-1 mRNA, Ezrin protein positive expression rate was 13.3% (2/15), 20.0% (3/15), breast cancer tissue of the MTA-1, Ezrin protein expression were 66.2% (53/80), 68.7% (55/80), the cancerous tissue MTA-1 mRNA, Ezrin protein positive expression rate compared to normal breast tissue, and the difference was statistically significant (P <0.05). (2) breast cancer, MTA-1 mRNA expression in stage Ⅲ, Ⅳ and Ⅰ, Ⅱ stage breast cancer group were 83.8% (26/31), 46.9% (23/49), with lymph node metastasis and those without lymph node metastasis of breast cancer, the positive expression rate was 86.9% (40/46), 41.1% (14/34), the difference was significant (P <0.05). With age, tumor size, pathological type of breast cancer patients positive expression rate was no significant difference (P> 0.05). (3) breast cancer, the positive expression rate of Ezrin protein in stage Ⅲ, Ⅳ and Ⅰ, Ⅱ of the breast cancer group were 87.0% (27/31), 48.9% (24/49), with lymph node metastasis group without lymph node metastasis of breast cancer, the positive expression rate was 89.1% (41/46), 47.0% (16/34), the difference was significant (P <0.05). At a different age, tumor size, pathological type of breast cancer patients positive expression rate was no significant difference (P> 0.05). (4) breast cancer, the MTA-1 mRNA expression of Ezrin protein was positively correlated (r = 0.634, P = 0.000). (5) breast cancer, MTA-1 3 / Ezrin 3 group of disease-free survival was significantly lower than the MTA- - / Ezrin - group (the x 2 = 7.610, P = 0.006). Conclusion: (1) breast cancer tissue MTA-1 mRNA expression of Ezrin protein expression was significantly higher than in normal breast tissue, suggesting that the MTA-1, Ezrin protein expression related to the occurrence of breast cancer. 2.MTA-1 mRNA expression of Ezrin protein and lymph node metastasis and clinical stage, has nothing to do with age, tumor size, histological type. MTA-1 mRNA, high Ezrin protein expression indicates that late tumor stage, lymph node metastasis potential. 3 joint detection MTA-1, Ezrin protein expression levels may provide for the judgment breast cancer prognosis basis.
