Abstract


Objective: ( 1 ) the establishment of rat donor liver cut animal models . ( 2 ) to explore the differences of rat donor liver cold preservation process of the superior vena cava lavage tumor necrosis factor TNF-α levels change over time . ( 3 ) to explore the vena cava lavage fluid in TNF-α levels and the relationship between the donor quality preimplantation evaluation of donor quality donor to seek a possible approach . Methods: The study in healthy male SD rats . ( 1 ) the establishment of a rat liver transplantation donor animal models cut cut donor 0-4 ℃ cold preservation . ( 2 ) the experimental animals were divided into seven groups , six animals in each group , respectively, when vena cava lavage specimens 1ml take 0,4,8,12,16,20,24 hours cold preservation measure liver function and TNF -α concentrations . Donor to do paraffin sections disease seized . (3) The statistical analysis of the experimental data . Results: ( 1) in the case of the same warm ischemia time , donor vena cava fluid of rats in liver function parameters , and TNF-α levels with cold preservation time increased . ( 2) donor liver donor liver biopsy found with obvious morphological changes save time extension . ( 3 ) donor vena cava fluid of rats in the correlation between the levels of TNF-α and ALT and AST indicators . Conclusion : ( 1) tumor necrosis factor in liver ischemia and reperfusion injury plays an important role , is one of the cytokines mediated mechanism of injury . (2) evaluation of donor livers quality indicators of liver function and pathological examination of the donor lavage have a certain value . ( 3) In this study confirmed the correlation between donor lavage TNF -α levels and liver function , and the difference between groups , the the cold shelf life donor lavage INF-α the level has the possibility of the evaluation of the graft quality . In fact, the value to be further experimental and clinical studies have confirmed .
