Objective: To study lupus nephritis (lupus nephritis, LN) in patients with urinary protein stimulated human proximal tubular epithelial cells (HK-2) NF-κB and its related inflammatory cytokines TNF-α, IL-6 expression and stimulation time correlation. Methods: 6 cases have been diagnosed as LN patients , and the clinical yet started adrenocorticotropic hormone and ( or ) cytotoxic therapy . LN patients 24 hours urine collection , urinary protein purification . Stimulated with 1mg/ml urinary protein serum-free medium for 24h HK-2 cells , stimulation time was 1h, 3h, 6h, 12h, 24h, 48h. Cultured in serum-free medium after 24h of HK-2 cells as a control , by reverse transcription polymerase reaction (RT-PCR) was observed at different time points urinary protein on HK-2 cells NF-κBmRNA, TNFαmRNA, IL-6mRNA expression . Results : 1mg/ml urinary protein can increase HK-2 cells NF-κBmRNA, TNFαmRNA, IL-6mRNA expression , with stimulation time expression increased , 48h peaked . Cultured with serum-free medium after 24h of HK-2 cells compared NF-κBmRNA, TNFαmRNA, IL-6mRNA expression , a statistically significant difference (P lt; 0.05). The correlation analysis, cell NF-κBmRNA, IL-6mRNA, TNFαRNA altered expression within 48h time-dependent manner , r values ??were 0.863,0.892,0.803 . P < 0.05 . Cell NF-κBmRNA with IL-6mRNA altered expression , TNFαRNA altered expression was positively correlated ( respectively r = 0.955, P <0.05 and r = 0.983, P <0.05). Conclusion: The patients with lupus nephritis 1mg/ml urine protein can stimulate HK-2 cells increased NF-κB, TNFα, IL-6mRNA expression , 48h within a time -dependent manner. NF-κB by upregulating the cytokine IL-6, TNFα, and thus mediates renal tubular interstitial inflammation and fibrosis.
