Establishment and Study of Human Oral Verrucous Carcinoma and the Expression of Survivin and Bcl-2 in Nude Mice Models

Abstract


Objective: Oral cancer nude modeling preliminary experimental study provides a useful exploration model for in-depth study Oral cancer occurrence , development and its evolution mechanism . MATERIALS AND METHODS: Oral cancer cancer tissue by tissue transplantation and cell suspension injection transplanted into Balb / c nude mice , the establishment of the human oral verrucous carcinoma in nude mice xenograft model (model I ) ; the recurrent subcutaneously in nude mice right after verrucous carcinoma associated with squamous cell carcinoma of neck metastasis by tissue transplantation method , to establish a relapse after verrucous carcinoma associated with squamous cell carcinoma of cervical lymph node metastasis in a nude mouse model ( model II) . Observed tumor growth characteristics , calculate the transplant success rate , the growth curve , and the transplanted tumor pathological and histological examination . Results: model modeling failed model primary transplant seven in eight nude mice to be successful, tumor formation rate of 87.5% ( 7 / 8 ) , tumor growth curves in accordance with the general laws of the Gompertz function curves . Transplanted tumors eventually necrosis , individual also appear subsided phenomenon . Subcutaneous tumor tumor spherical or oval , smooth surface , encapsulated , nodular protrusions clear boundaries with the surrounding tissue , a good activity , some tumors showed concentric necrosis , some tumor clearly visible on the surface of the capillary blood vessels , the surface of red meat , sections were fish-like quality and tough , and some white bean dregs like . Light microscope transplanted tumor cells into polygonal atypia , nuclear cytoplasm ratio increases . Nuclear oval , round , ranging in size , mitotic common, and see pathological karyokinesis . Cancer cells arranged in the nested sheets or cords during visible keratin pearls . Liver , lung, kidney and no metastasis , the forelimb armpit and groin lymph node reactive hyperplasia . Transplanted tumor of Survivin, Bcl -2 protein immunohistochemical staining and human tumor difference was not significant ( P > 0.05 ) . Conclusion: 1 . Establish a relapse after verrucous carcinoma with squamous cell carcinoma of cervical lymph node metastases in nude mice primary xenograft model to be successful , and laid a solid foundation for the next step to establish a stable xenograft model . 2 people with recurrent verrucous carcinoma associated with squamous cell cervical lymph node metastasis in primary transplant tumor retain biological characteristics of human squamous cell carcinoma , and similar to the histological features of the primary tumor . 3.Survivin and Bcl-2 expression in transplanted tumors and well-differentiated squamous cell carcinoma , the difference was not statistically significant .
