Objective: To study evodiamine regulating the biological function of DC, DC-based anti-inflammatory immunotherapy with independent intellectual property rights and lay a foundation for the development. The search for new anti-inflammatory drugs, traditional Chinese medicine and exploration and interpretation of its pharmacological effect has become a trend. Evodiamine Chinese medicine Evodia alkaloids, with a variety of biological activity, in recent years, studies have shown that it has significant anti-inflammatory activity. Dendritic cells (DC) as the most powerful professional antigen-presenting cells in the start-up and connection of innate immunity and acquired immunity plays a vital role. In the maturation process of DC antigen uptake, antigen presentation, migration, phenotypic changes and cytokine secretion function affects the entire immune system, Evodiamine DC function has not been reported so far, therefore, evodiamine on DC The variation of the torsion inflammatory response caused by immune disorders has important significance. Immature DC (iDC) cells: separation of Balb / c mice bone marrow cells induced by GM-CSF in vitro cultured for 10 days treatment on various factors and is divided into: control group (I), evodiamine group (Evo) (II ), endotoxin (LPS) group (III), Evo LPS (Ⅳ). Evodiamine treatment of mouse bone marrow-derived DC stimulation of proliferation of non-adherent cells in the mixed lymphocyte reaction assay. ELISA test for detection of mouse bone marrow-derived DC and evodiamine in vitro co-culture at different times during the secretion of IL-10 and IL-12 levels. 3 flow cytometry evodiamine treated mouse bone marrow-derived DC surface molecules of CD11c, MHC Ⅱ, of CD40, CD80 and CD86. 4. Gene chip technique evodiamine impact on the the DC functional regulation of gene expression. Results: Compared with the control group, the endotoxin group and evodiamine group absorbance value was significantly higher (P lt; 0.05); the endotoxin group Evodiamine evodiamine group compared evodiamine endotoxin group absorbance values significantly higher than the evodiamine group (P lt; 0.05); evodiamine group endotoxin group compared with the endotoxin, evodiamine group absorbance significantly higher endotoxin group (P lt; 0.05). Evodiamine treatment before and after iDC cytokine secretion levels are very low, below the detection sensitivity; mDC evodiamine treatment for 24h, 48h after IL-12 (P40), IL-12 (P70) and IL-10 treatment Before comparison, no significant change (P gt; 0.05). 3 the endotoxin group with the control group compared to that of MHC II, CD40, CD80 expression was significantly higher (P lt; 0.01, P lt; 0.05); the endotoxin group Evodiamine with Evodiamine group compared, Evodiamine endotoxin Group of MHC II, CD40, CD80 and CD86 was significantly higher than the of evodiamine group (P lt; 0.01, P lt; 0.05); the evodiamine group and the control group compared with of CD80, CD86 (P lt; 0.05) expression significantly down-regulated. 4. Ⅱ / Ⅰ raised ≥ 2-fold gene 7, down ≥ 2-fold gene 10; 37 ≥ 2-fold gene III / Ⅰ raised, lowered ≥ 2-fold gene 12; Ⅳ / II raised 46 ≥ 2-fold gene ≥ 2-fold, down seven genes; Ⅳ / III raised 24 ≥ 2-fold gene down ≥ 2 genes 2. Conclusion: 1. Evodiamine not only promoted iDC antigen-presenting function, while also promoting the mDC antigen-presenting function. Evodiamine iDC, mDC cytokine IL-10, IL-12 secretion function. Evodiamine inhibit iDC part of the co-stimulatory molecule expression of MHC Ⅱ molecules on mDC costimulatory molecules had no effect. 4 the evodiamine DC role involving multiple gene expression and regulation of these genes control and affect the function of DC differentiation and maturation find drug targets for further clues.
