Abstract


Tomato late blight[Phytophthora infestans (Mont.) De Bary] is a devastating plant disease all over the world. The genetic structures and composition changes of the pathegen are related closely with the occurrence and prevalence of the disease and have a direct impact on it.The identification and research on the phenotype and genotype of the pathegen have been carried out after the outbreak and epidemic of the disease became more and more frequent in many countries which had main tomato cultivation since the 1980s.A complete set of technology system had been established and the systematic research results were obtained in the foreign countries.Jiangsu province is located in the Yangtze River Delta region,which is one of the important growing areas of tomato in China.Because of the cross-linking of the geography,the transition of the climate as well as of the diversity of cultivation,the species of the pathegen may be complicated and varied.Tomato late blight will become more and more serious which will cause much more serious harm to its cultivation.So,there is an urgent need to carry out the studies related.In this experiment, extensive sampling was carried out in tomato-growing areas from Jiangsu province and its surrounding areas.Strains of Phytophthora infestans were isolated successfully,and the related identification and analysis of phenotype and genotype were also done.The genetic composition and distribution of the pathegen as well as mainstream groups were clear preliminarily in these areas,which laid the foundation for the production of high-effective,low-poisonous anti-fungal agent, breeding resistance varieties against tomato late blight. The main findings are as follows:(i).Use selective medium and choose the pathogenesis diseased leaves and neck that have typical symptoms to separate,the law of organization separation has the highest rate of separation.Single pathogen alternaria purification is the best purification method.91 strains of Phytophthora infestans were obtained successfully.(ii).The mating type of 91 strains of Phytophthora infestans were identified,all were A1 mating type;no A2 mating type strains were found.It is concluded that the sexual reproduction of the pathegen did not happen under natural conditions in the tomato-growing areas of Jiangsu province and its surrounding areas. (iii).mtDNA haplotypes were identified on 91 strains of Phytophthora infestans, the results showed that:there were two mtDNA haplotypes:Ib and IIb.The number of IIb types of the strains is 86,which account for 94.5% of the strains tested,and have been found in all areas of the collection,the IIb type is the advantage mtDNA haplotypes in the sampling areas.The Ib type is only found in Xuzhou Jiangsu and Hexian Anhui with small quantities.Analysed comprehensively,the "new" group of Phytophthora infestans is the main one in the areas above.(iv).Genotypes of 6-glucose phosphate isomerase (Gpi) and peptidase (Pep) on 91 strains of Phytophthora infestans were identified and analysed.The results showed that there were four Gpi types: 100/100/111、86/100/111、86/100 and 100/100;two Pep types:100/100 and 92/100.A total of six allozyme genotypes were found: Gpi(100/100)/Pep(92/100)、Gpi(100/100/111)/Pep(100/100)、Gpi(86/100/111)/Pep (100/100)、Gpi(86/100)/Pep(92/100)、Gpi(100/100)/Pep(100/100)、Gpi(100/100/111) /Pep(92/100).The type of Gpi(100/100/111)/Pep(100/100) distributed most widely in the areas of sample collection with the largest number,and was the dominant groups.The other five types differ obviously between different regions. Xuzhou Jiangsu is the most complex area, followed by Hexian Anhui and Qingdao Shandong.The allozyme genotypes of Phytophthora infestans in Suqian、Nanjing、Zhenjiang、Yangzhou and Nantong were relatively simple.The allozyme genotypes of the pathegen groups are relatively complex with high level of diversity in the sample collection areas.
