Abstract


Gene imprinting (genomic imprinting) newly discovered genetic rule in recent years, with the growth and development of the embryo, a rare genetic disease and cancer are closely linked, and phenomenon explains many Mendelian law can not match, its significance is gradually also becoming more in-depth understanding of research. The traditional Mendelian law: embryonic inherited from the father and the mother of two copies of that allele (alleles) have an equal opportunity to express their parental sources unrelated, but in recent years found that a small part of the gene's expression depends on these genes from the father or from the mother this allele expression depends on the parental origin of the phenomenon that is genetic imprinting, monoallelic expression of certain genes was pro source dependence, and the other allele did not express or expression of extremely weak. This phenomenon genes called imprinted genes (imprinted gene). Gene imprinting in human genetic diseases, in particular the role in tumorigenesis is attracting more and more attention. A growing number of studies have shown that abnormal imprinted genes, including loss of imprinting (loss of imprinting, LOI) biallelic expression plays a role in tumorigenesis. H19 gene is located on human chromosome 11, and expression of paternally imprinted maternal only expression of RNA, are not translated into proteins, and its function is unclear, some studies have shown that the growth inhibition plays in tumorigenesis. Many studies have found that the the H19 gene not only in children embryonic malignancies and occurs in many malignant tumors in the adult LOI. Long QT intron transcript 1 (long QT intronic transcript 1, LIT1) is positioned in the human chromosome 11, the expression of maternal imprinting, the parent source is the antisense transcripts of the the KvLQT1 gene paternal allele, and The p57KIP2 gene imprinting maintain. Also found in some malignant LOI LIT1 gene. Mesodermal specific transcript (also known as the parent source expressed genes -1) (Mesoderm-specific transcript, MEST) located on human chromosome 7q32, a maternal imprinted paternally expressed gene, has been reported in some malignant tumors its overexpression and its occurrence LOI related. About three imprinted gene imprinting status change and lung cancer, is rarely reported abroad, the domestic has not been reported. Objective: To investigate the H19 imprinted genes in human lung cancer tissue, LIT1 and MEST imprinted deletion mechanisms and possible role in lung cancer. Methods: PCR technology combined with restriction fragment length polymorphism analysis (restriction fragment length polymorphism, RFLP) screened with 32 cases of human lung cancer tissue and corresponding adjacent tissues H19 LIT1 and MEST gene heterozygosity cases , and then using the RT-PCR-RFLP assay H19 the LIT1, and MEST blot state. Results: In 32 cases of group of H19, LIT1 and MEST gene heterozygous individuals were 14, 4 and 13 heterozygosity ratios were 43.8% (14/32), 12.5% ??(4 / 32) and 40.6% (13/32); H19 and MEST in lung cancer The biallelic expression rates were 28.6% (4/14) and 53.8% (7/13), while in the corresponding adjacent tissues in dual allelic expression rates are 0, the difference was statistically significant (P lt; 0.05), LIT1 tissues in the lung tissue and corresponding paraneoplastic biallelic expression rates were 25% (1/4) and 0, the difference was not statistically significant (P gt; 0.05). Conclusion: The lung tissue can be detected the occurrence of an LOI of H19 and MEST gene H19 and MEST gene LOI may be involved in the process of lung cancer.
