Relationship between Apoptosis with MCM4 and CDC6, CDK1, TFF2 and HPV16/18 Infection in Uterine Cervical Carcinogenesis

Abstract


Objective: To study MCM4, of CDC6, CDK1, TFF2 expression in cervical cancer and its Human Papillomavirus infection, in order to investigate the mechanism of the occurrence of cervical cancer, and look for new molecular markers is conducive to the diagnosis of cervical cancer, to guide the diagnosis and prognosis. Methods: The immunohistochemical method in 50 cases of cervical squamous cell carcinoma, 20 cases of CIN I, 20 cases of CIN II - III, 20 cases of normal cervical tissue the MCM4, CDC6, CDK1, TFF2 of detection, while using the PCR technique HPV16/18 infection . Statistical methods of experimental data: count data using the chi-square test, Fisher's exact test; hierarchical data using non-parametric rank sum test; correlation test using Spearman's rank correlation analysis of data. Results: (1) in cervical cancer MCM4, CDC6, CDK1 positive rate in CIN and normal cervical tissue, the difference was significant (P lt; 0.05), and MCM4, CDC6, CDK1 positive expression rate is correlated with pathological grading , the difference was significant (P lt; 0.05), MCM4, the CDC6 positive expression rate of lymph node metastasis (P lt; 0.05); MCM4 in cervical cancer, the positive expression rate of CDC6 and CDK1 age group and clinical stage (P gt; 0.05) In cervical cancer in a TFF2 the positive rate lower than the CIN and normal cervical tissue, the difference was significant (P lt; 0.05), the positive expression rate of the age group (P lt; 0.05), the positive expression rate of clinical stage, histological grade, lymph node metastasis (P gt; 0.05) (2) Expression of Human Papillomavirus Type 16 and 18 in normal cervical tissue, CIN and cervical cancer, the positive rate is gradually increased, the difference was significant (P lt; 0.05), but with clinical stage, histological grade, lymph node metastasis, independent of the age group ( p gt; 0.05) (3) MCM4, CDK1 \u0026 CDC6 expression was positively correlated (rs = 0.390, P lt; 0.05; rs = 0.529, P lt; 0.05); MCM4 and CDC6 HPV16/18, infection (rs = 0.634, P lt; 0.05; rs = 0.386, P lt; 0.05); TFF2 with MCM4, CDC6, HPV16/18 express negative correlation (rs = -0.375, P lt; 0.05; rs = -0.356, P lt; 0.05; rs = -0.500, P lt; 0.05). Conclusion: The cervical cancer tissues MCM4, CDC6, TFF2 expression of the change may be infected with HPV16/18, and the interaction, the combined effect of the occurrence of the development of CIN and cervical cancer. Human Papillomavirus is an important factor leading to increased susceptibility to cervical cancer, but not the only factor. MCM4 the CDC6, of CDK1 abnormal expression of cervical cancer malignant degree can be used as screening for cervical cancer, the favorable prognostic indicators.
