Objective To investigate the mononuclear cells (MNC) count as a predictor of allogeneic peripheral blood stem cell transplantation hematopoietic reconstitution indicators is feasible. Methods from January 2000 to December 2007, 98 patients (60 males and 38 females, median age 30 years) in the transplant center allogeneic peripheral blood stem cell transplantation. 98 sibling donors (50 males and 48 females, median age 28 years) to accept a unified mobilization program: rhG-CSF5μg · kg -1 sup>, subcutaneously, twice daily interval of 12 hours of administration, on the morning of the fifth day after 2 hours of administration Cobe Spectra blood cell separator to MNC procedures PBSC collection. 98 cases are divided into MNC group (61 cases) and CD34 sup> cells group (37 cases), MNC group to MNC ≥ 4 × 10 8 sup> / Kg for the acquisition target, CD34 < sup> sup> cells group to CD34 sup> cell count ≥ 4 × 10 6 sup> / Kg for the acquisition target. Comparison of two counts indicators engraftment and donor acquisition times of. Results ① MNC group recipients enter MNC median of 6.75 (3.03 ~ 18.57) × 10 8 sup> / Kg, CD34 sup> cells group recipients input CD34 sup> cells median 5.05 (2.06 ~ 17.23) × 10 6 sup> / Kg. ② MNC group and CD34 sup> cells group recipient hematopoietic reconstitution rate was 100%; groups ANC> 0.5 × 10 9 sup> / L, the median time was 11d (9d ~ 17d) and 11d (9d ~ 27d), no statistically significant difference (P> 0.05); groups PLT> 20 × 10 9 sup> / L, the median time was 12d (9d ~ 71d) and 12d (9d ~ 35d), no statistically significant difference (P> 0.05); ③ donor an acquisition rate: MNC group and CD34 sup> cells group were 100% and 37.84%, the difference There was statistically significant (P <0.05); ④ MNC group with mismatched HLA identical sibling transplant recipients ANC> 0.5 × 10 9 sup> / L, the median time was 11d (9d ~ 16d) and 12d (10d ~ 17d), the difference was not statistically significant (P> 0.05); PLT> 20 × 10 9 sup> / L, the median time was 12d (9d ~ 71d) and 14d (10d ~ 22d), the difference was not statistically significant (P> 0.05). Conclusion MNC as hematopoietic stem / progenitor cell levels count indicators, not only can reliably predict sibling donor peripheral blood stem cell transplantation (including HLA-identical and mismatched transplants) after hematopoietic reconstitution, the engraftment rate and speed of engraftment with CD34 < sup> sup> cells is comparable to, but once its donor acquisition rates (100%) was significantly higher than the latter (37.8%), the results indicate that MNC count can replace CD34 sup> independent as hematopoietic stem cells / progenitor cell levels indicators.
