Abstract


Objective: To study the traditional Chinese medicine monomer psoralen OB-OC co-culture system , the proliferation of osteoblasts (Osteoblast, OB) , alkaline phosphatase ( Alkaline phosphatase , ALP) activity of psoralen on OB interleukin -11 ( interleukin -11 , IL -11 ) mRNA expression of affect , to explore whether the psoralen pathway has a role in the prevention and treatment of osteoporosis (Osteoporosis, OP) and its possible . Method: 1 . Taken 1d old SD rats separation OB take 5d old SD rats separation osteoclasts ( Osteoclast OC) , the establishment of OB-OC co-culture system . 2 . MTT assay was used to detect the concentration of each drug on the OB-OC Sterile OB proliferation system , to determine the optimal concentration of the drug . 3 p-nitrophenyl phosphate (P-nitrophenyl phosphate, PNPP) detection OB ALP activity ; Alizarin Red staining OB mineralization . 4.RT-PCR method to detect of psoralen impact of of IL-11mRNA expression . Results: 1. Coomassie blue assay protein diffusion case, show that within 8h , protein from OB chamber through the semi-permeable membrane dispersion to OC room . 2.OB co-cultured with OC , OD values ??than when cultured alone significantly improved (P <0.05). 3.OB-OC OB ALP activity in the co-culture system OB ( P <0.05 ) higher than training alone , the number of mineralized nodules OB ( P <0.05 ) was significantly higher than that cultured alone . 4 psoralen concentration 5μmol / L the OB OD values ??(P <0.05), ALP activity was higher than that in the control group ( P <0.05 ) . 5. The psoralen intervention OB-OC the total sterility system OB the IL-11mRNA of expression level higher than that of the control group ( P <0.05 ) . Conclusion : vitro OB OB-OC co-culture system in the proliferation , differentiation and mineralization ability were strong . Psoralen concentration was 5μmol / L , significantly promote OB proliferation , increase ALP activity . The psoralen improve the co-culture system OB the IL-11mRNA of expression levels .
