[Objective] In this study, the use of the characteristics of the pH-dependent drug delivery systems , the preparation of berberine hydrochloride colon controlled release coated tablets , in order to achieve the purpose of the colon targeted drug delivery , improve its antimicrobial efficacy . [Method] , selected on the basis of the orthogonal test conducive to disintegration and dissolution of the tablet core fillers , optimization filler proportion investigated disintegrants, pressure and other factors , the immediate release and having a good expandable tablet core ; acrylic resin II number , Ⅲ number according to different proportions for the coating material , so that the drug delivery system to adjust the coating thickness in a hydrochloric acid solution of 0.1mol / l , pH 6.8 , pH 7.4 phosphate buffer solution of simulated stomach, small intestine, colon tract environment through the stomach and small intestine environments and direct colonic release . Evaluate the quality of the tablet and stability study . [Results] The the best tablet cores prescription and best coating the prescription ; obtained berberine colon targeting controlled-release tablets in the pH6.8 buffer 3h substantially insoluble , at pH7.4 buffer 1h dissolved more than 75% , in line with the the tablet quality standards two appendices of the 2005 edition of \[Conclusion] The preparation process is feasible , colon targeted controlled release effect , reliable and accurate method of quality control , the validity is tentatively scheduled for 2 years .
