Abstract


White Spot Syndrome pathogen as a body embedded class baculovirus , this virus is named for the white spot syndrome virus ( White spot syndrome virus of WSSV ) . Disease prevalence and mortality are high , and the Host range widely , causing huge economic losses to the shrimp industry . In this study the material suffering Procambarus clarkii WSSV disease , return a sense of healthy crayfish , the establishment of a stable white spot syndrome virus back to a sense of system ; designed and in accordance with the published WSSV genome sequence (GenBank accession number AF332093) primer by PCR amplification of an unknown gene sequences of WSSV ORF 332 , sequence analysis results show that the the WSSV gene pool of the sequences in GenBank corresponding 98 % sequence homology , recognized as crayfish WSSV gene fragment Macrobrachium rosenbergii of WSSV ; another detection by PCR method , results show that the natural host of M. rosenbergii is also WSSV ; M. rosenbergii of WSSV ORF 234 cloned in a prokaryotic system expressed by SDS-PAGE and WesternBlot detect expression product , the results show the the ORF 234 can properly express the protein , Procambarus clarkii WSSV amplified with M. rosenbergii WSSV ORF 234 similar specific bands . WSSV great harm to the host domain widely , accurate and sensitive diagnosis of WSSV has a positive meaning for aquaculture production . This study on two of ORF region in the WSSV genome sequence analysis, two sets for LAMP -specific amplification primers were designed , and the results show that the two sets of primers can be accurately amplified expected strip temperature is an important factor affecting the amplification results . ORF 234 fragments amplified LAMP sensitivity to nearly ten thousand times higher than the PCR amplification , and does not have a false positive , amplification can be completed within 1.5 h , with time provincial high sensitivity and no false positive required simple equipment , etc. , to promote the use of good prospects . The study also selected WSSV gene fragment , codon preference analysis results show : the sequence codon adaptation index (CAI) low synonymous codon codon coefficient (FOP) is close to 0.4 , codon bias index (CBI) majority is negative , indicating that the best codon in these areas is less than the number of expectations , suggesting that the virus codon usage is low , this is perhaps one of the reasons why the virus host domain wide .
