Abstract


[Objective] To study the beta 1,4-N- acetyl galactose amino transferase (GalNAc T ) mRNA in early patients with lung cancer , peripheral blood and lymph nodes of expression as well as among explore it as tumor markers feasibility of lung micrometastases and its clinical significance . 【Methods】 by using nested reverse transcription polymerase chain reaction (nested RT-PCR) technology , detection by the surgical treatment of 35 cases of Ⅰ Phase II lung cancer patient specimens (cancerous tissue, lymph nodes and peripheral blood ) GalNAc T mRNA expression ; choose the 12 cases of benign lung disease patients surgical specimens and 10 healthy volunteers blood samples as controls ; 247 regional lymph node micrometastases and surgical resection and conventional pathological examination for comparison . [Results] The pathologic examination confirmed 35 cases of stage I , Phase II lung cancer patients , GalNAc T mRNA was positive in 22 cases ( 62.86% ) , and 12 patients with benign lung tumors were negative groups there are significant differences ( P LT ; 0.01 ) . Peripheral blood samples of patients with lung cancer in 8 cases ( 22.86% ) positive for GalNAc T mRNA , 12 benign tumor patients and 10 healthy volunteers were negative group difference ( P lt; 0.05 ) . GalNAc T mRNA expression in lung cancer patients with positive rates with gender , age, smoking history , tumor type and degree of differentiation was no significant relationship ( P gt ; 0.05 ), with TNM stage about (P lt; 0.05). 247 surgical removal of regional lymph nodes , RT-PCR method detected 113 (45.75 %) lymph node micrometastases , only 61 (24.70 %) of the routine pathological examination of lymph node metastasis , the difference between the two was analytical significance (P lt; 0.05). [Conclusion] molecular biology methods to diagnose lung cancer lymph node micrometastasis more sensitive than conventional histopathological methods . Using nested RT-PCR method to detect GalNAc T mRNA expression in peripheral blood higher specificity , and closely associated with lymph node micrometastasis . GalNAc T may be potential lung cancer tumor markers , can be used as a reference for the early diagnosis of lung cancer patients with micrometastases , provide important theoretical basis for the selection of treatment options and assess prognosis .
