Abstract


Objective: The specific change of systemic scleroderma (systemic scleroderma, SSc) is an extracellular matrix (extracellular matrix, ECM) excessive deposition in the skin and internal organs, causing autoimmune diseases. Its characteristic change is excessive collagen production. Matrix metalloproteinases (matrix metalloproteinases, MMPs) are a group of selective degradation of extracellular matrix endopeptidases and their inhibitors (Tissue inhibitor of metalloproteinases, TIMPs) is an important factor in regulating MMPs. MMPs and TIMPs play an important role in maintaining the ECM homeostasis. Matrix metalloproteinase 9 (matrix metalloproteinase-9, MMP-9) and their inhibitors (tissue inhibitor of metalloproteinase-1, TIMP-1) play an important role in tissue inflammation and fibrosis process. Therefore, we examined the systemic scleroderma and localized scleroderma, MMP-9 and TIMP-1 investigate the expression of MMP-9 and TIMP-1 in systemic scleroderma and localized scleroderma (localized scleroderma LSc) possible role in the pathogenesis, provide the basis for its pathogenesis, immunology. Methods: 21 cases of SSc and 21 cases LSc-skin lesions of patients for the experimental group, 10 cases of surgical resection of normal skin for the control group, with the immunohistochemical the SP assay detected SSc and LSc lesions of MMP-9 and TIMP-1 The expression and compared with normal skin. MMP-9 and TIMP-1 expression results of immunohistochemistry applications HPIAS-1000 high-resolution color the pathology graphic analysis system, respectively, measured MMP-9 and TIMP-1 coloring mean optical density value, indicating that the expression of the strength of the ingredients . The two groups were compared using the t test, completed application SPSS11.0 statistical software. Results: 1, the expression of MMP-9: 10 cases of normal skin in the basal layer were positive for the expression of the strong expression of three cases, four cases of positive expression, four cases of weak expression; acantholysis strong positive expression 1 were positive, weak positive expression, two cases with negative expression. In the granular layer two cases weakly positive expression, negative expression of eight cases. 21 patients with SSc group in the basal layer were positive for the expression of the strong expression of six cases, 13 were positive, two cases of weak positive expression; 11 cases acantholysis weakly positive expression, more than 10 cases of negative expression. Of 21 cases LSc patient group in the basal layer were positive for the expression of the strong expression of three cases, 11 were positive, seven cases of weak positive expression; acantholysis positive expression, four cases of weak expression. T test for two sets of data were analyzed using SPSS11.0 statistical software: MMP-9 expression average luminosity values ??lower in SSc group than in the control group (P lt; 0.05), compared with normal control LSc group reduced but between the two groups no significant difference (P gt; 0.05). 2, TIMP-1 expression: the control group, 10 cases of normal skin in the basal layer of strong positive expression, two cases of positive expression, five cases of weak expression; acantholysis strong expression of the two cases weakly positive expression. In the granular layer and stratum corneum were negative. Strong positive expression in 18 cases of 21 cases LSc patient group, 3 patients with positive expression; nine cases acantholysis weak expression. Weak expression in the granular layer two cases, I was negative expression. The strong positive expression in 19 cases of 21 patients with SSc group, 2 patients with positive expression; acantholysis eight cases of strong positive expression, 6 were positive, 6 cases weakly positive expression. Patients with weak expression in the granular layer, the remainder were negative. TIMP-1 expression in of SSc and LSc lesions were significantly higher than that of the control group, the differences were statistically significant (P lt; 0.05) among the three groups. Conclusion: MMP-9 and TIMP-1 may be related to the pathogenesis of systemic scleroderma related.
