The Effect of Low-dose Radiation on PCNA and EGFR of Tumor in S180 Sarcoma-bearing-mice

Abstract


Objective To observe the low-dose total body irradiation of mice general and its inhibition effects of low dose total body irradiation of mice bearing S180 sarcoma tumor tissue PCNA, EGFR impact , to further explore the mechanism of action of low-dose radiation . Methods Kunming mice were inoculated subcutaneously S180 sarcoma cells ( 12 days) after the formation of the tumor, and the mice were randomly divided into control group (N) and low - dose radiation group ( LDR ) . Of the the LDR group of mice given disposable 75mGY Co60 ray body irradiation , sham group were given no false -ray irradiation . Mice generally observe the sham group is put to death after irradiation 6h LDR group were irradiated after 6h (LDR-6h group ) 12h ( LDR -12h group ) , 24h (LDR - 24h group ), 48h (LDR- 48h group ) , 72h (LDR - 72h group ) were sacrificed . Respectively, the tumor said the weight of tumor , histological analysis and immunohistochemical staining of tumor tissue , proliferating cell nuclear antigen ( PCNA ) , epidermal growth factor receptor (EGFR) expression . Results of the LDR each group tumor weight with no significant difference between the sham group (N) ; the LDR group PCNA expression irradiation decreased with time after , LDR-24h group , LDR-48h group , LDR - 72h group and the sham group are statistically significant difference ( p <0.05 ) , the the LDR - 48h group with the the LDR-24h group compare PCNA expression decreased statistically significant ( p <0.05 ) ; the LDR group of EGFR expression began to decrease after irradiation is rendered , LDR-12h group sham group was statistically significant (p <0.05), LDR-24h group was the lowest . Conclusion Low dose total body irradiation ( the mice are irradiated with 75mGy ) 72 hours can reduce PCNA expression in mice bearing S180 sarcoma tumor tissue , inhibit tumor growth ; 24 hours of low-dose total body irradiation ( the mice are irradiated with 75mGy ) mice bearing S180 sarcoma tumor tissue can be cut EGFR expression affect tumor cell signaling .
