The Effect of Imidapril and Valsartan on Inhibiting the Develepment of Atherosclerosis and Its Mechanism

Abstract


Objective:To study the effect of imidapril and valsartan on the expression of CDK4、VCAM-1 and p15 mRNA in rabbit aorta,and investigate the anti-atherogenic mechanism of them.Methods:Fourty rabbits were randomly divided into 4 groups:the control group,high cholesterol group,high cholesterol+imidapril group and high cholesterol+valsartan group,ten rabbits in each group.Blood and artery samples were collected at the end of 8 weeks for examination of serum lipid levels,the expression of CDK4、VCAM-1 detected by immunohistochemistry and the expression of p15 mRNA detected by RT-PCR.Results:Rabbits fed with cholesterol-rich diet showed higher serum lipid levels(q1=23.34,q2=23.06,q3=22.98,P<0.01) than those fed with normal diet.Serum lipid level of Rabbits fed with cholesterol-rich diet showed no difference(q1=1.74, q2=0.81,P>0.05).The ratio of lesion of intimal area of imidapril treated group and valsartan treated group reduced significantly than those of high cholesterol group (q1=65.60,q2=65.68,P<0.01).The expression of CDK4 and VCAM-1 of imidapril treated group and valsartan treated group decreased significantly than those of high cholesterol group(qCDK41)=16.11,qCDK42=17.9,qVCAM-11=4.42,qVCAM-12=5.93,P<0.01).while the expression of p 15 mRNA of imidapril treated group and valsartan treated group is higher than those of high cholesterol group(q1=4.38,q2=5.78,P<0.01).But the expression of these sign shows no difference between imidapril treated group and valsartan treated group(P>0.05).Conclusion Valsartan,imidapril can inhibit atherosclerotic progression.The mechanism may be related to inhibition of inflammation and the proliferation of vascular smooth muscle cells in atherosclerosis.
