Objective To investigate the self- rat peripheral blood endothelial progenitor cells (endothelial progenitorcells EPCS) approach for stem cell transplantation in the treatment of ischemic myocardial injury provide new sources of non-invasive cells . Approach taken by continuous subcutaneous rat granulocyte colony -stimulating factor (Rat GranulocyteColony Stimulating Factor rG-CSF) seven adult male Sprague-Dawley (SD) rats with blood , the application of Ficoll (Ficoll 400) density gradient centrifugation extraction rat peripheral blood mononuclear cells , and then to 20% sugar IMDM, fetal bovine serum, horse serum , basic fibroblast growth factor (basic fibroblast growth factor bFGF) and vascular endothelial growth factor (vascular endothelial growth factor VEGF) such as a mixture of complete medium , and cultured in vitro amplification , the resulting cells were cultured CD31, CD34, Flk-1 and vW F immunofluorescence staining and detecting the binding UEA-1, ac-LDL uptake ability . Results The cultured cells obtained by CD31, CD34, Flk-1 and vW F immunofluorescence staining and determination of binding UEA-1, the ability to ac-LDL uptake , were positive , confirming the endothelial precursor cells (EPC). Conclusions from rat neutrophils mobilized peripheral blood mononuclear cells were cultured in vitro , successfully endothelial precursor cells (EPC), endothelial progenitor cells (EPC) transplantation therapy is expected to become a new ischemic myocardial injury non-invasive cell source .
