Abstract


The study was conducted to investigate the effects ofα-Ketoglutarate on growth performance, intestinal function and plasma parameters in piglets with Lipopolysaccharide (LPS) stimulated repeatedly.Twenty four healthy crossbred (Duroc×Landrace×Largewhite) weanling piglets (22±2 days-old, 6.79±0.32 kg BW) were randomly allocated into four treatments. There are six replicates per treatment with one piglet each replicate. Piglets were assigned to a 2×2 factorial arrangements with LPS challenge (with or without) and dietary supplementation of AKG (0% or 1%) as the main factors.A trial period of 16 d. Piglets in the stress group were injected intraperitoneally with 100μg/kg?BW LPS at d 10, 13 and 15, and piglets in the no-stress group were injected with normal saline at the same dose. At day 10 and 15,Blood samples were collected from precava vein after 3 h of LPS or physiological saline injection.At day 16, all piglets were killed after injected with 50mg/kg?BW pentobarbital sodium 15 minnuts later, Scraped intestinal mucosa, thrown into the quick-frozen in liquid nitrogen and then turn preserved in refrigerator at -80℃until analysis. The results show that:(1) The growth performance of different groups no significant difference in the test from d 1~9 (no-stress time) (P>0.05); ADG was significantly decreased under repeated LPS stimulation (P<0.05)(stress time) and AKG have interaction with LPS stimulate repeatedly. LPS stimulation increased several piglets plasma alanine aminotransferase (P<0.01), total protein (P<0.05), globulin (P<0.01) levels, reduced albumin/globulin (P<0.01). AKG decreased plasma alanine aminotransferase levels (P<0.05) in the day 15. AKG of the total protein, globulin and albumin/globulin interacted with LPS stimulation (P<0.05). The results showed that diets supplemented with 1% AKG repeated LPS stimulation can ease the daily gain decreased and plasma biochemical changes.(2) LPS stimulation of plasma DAO activity several times higher (P<0.05), jejunal mucosal DAO activity and plasma D-xylose content decreased (P<0.05); and AKG and LPS on plasma and jejunal mucosa there was an interaction DAO (P<0.05), plasma D-xylose content of the possible interaction (P=0.097). LPS stimulation led to multiple jejunal mucosal tight junction proteins Claudin-1 expression decreased relative (P<0.05), AKG increased the relative jejunal mucosa Claudin-1 expression. The results showed that diets supplemented with 1% AKG can be mitigated to some extent repeated LPS stimulation decreased intestinal barrier and absorption functions.(3) for the first time and repeated LPS stimulation have led to plasma malondialdehyde (MDA) content increased (P<0.05); AKG and a series of LPS stimulation on the possible interaction of MDA (P=0.094); the first LPS stimulation the plasma superoxide dismutase (SOD) activity was significantly lower (P<0.05), AKG and repeated LPS stimulation there was an interaction of the SOD activity (P <0.05). The results showed that diets supplemented with 1% AKG have a certain weakening of LPS stimulation induced oxidation.In conclusion, in normal physiological conditions (not injected with LPS), the diet supplemented with 1% AKG no effect on piglet. LPS stimulation led to many Intestinal barrier function and decreased absorption, increased free radicals, diets supplemented with 1% AKG can alleviate the LPS stimulation led to slow growth of piglets, the mechanism is enhanced antioxidant, increased intestinal absorption, and barrier function.
