Xinjiang is China's largest sheep breeds, the most complex genetic quality of the provinces, and a number of distinctive local varieties bred in the long-term production practice, these varieties of precious wealth of the world's animal resource development and utilization. Kazak sheep has a large body precocious, strong resistance, adaptability, and other good traits; Altay sheep is a sheep Kazakh excellent groups with big physique, and lamb growth and development, increase fat fast, high-fat content of the meat production ; characteristics Bashibai sheep with precocious puberty, resistance to crude feed, adaptable coat is brownish red, more fluff, hair less dry out and die. These local varieties is an important species to protect biodiversity resources is the basis for sustainable development of the sheep industry in China. This study in the country for the first time the use of AFLP markers from the molecular level to clarify the Kazak sheep, Altay sheep, Bashibai the the polymorphism of sheep breeds resources, to provide a scientific basis for the protection and rational use of local varieties. This experiment using AFLP markers Kazak sheep, the Altay sheep Bashibai the sheep genetic analysis of the statistics of the total number of bands for each primer combinations detected in all varieties and number of polymorphic bands calculated three local varieties of sheep genetic similarity coefficient and genetic distance, which was constructed UPGMA cluster diagram. The study results are as follows: Compare the boiling water bath method, the quality of the extraction the Miller salting-out method, CTAB method, phenol method and improved sheep genomic DNA, results boiling water bath purity and concentration of the extracted DNA worst effect improved phenol The extraction method and the the Miller salting-out method, CTAB method compared extracted DNA product purity and concentration of a significant difference (P lt; 0.05), suitable for AFLP and experimental requirements DNA template. 2 In this study, a total of 64 pairs of MseI and EcoRI primer combinations repeated screening, which are amplified by the strip many and polymorphism high percentage with rich, clear bands of primer combinations, 11 pairs (E01/M02 , E01/M07, E02/M02, E04/M01, E05/M05, E05/M08, E06/M02, E06/M04, E06/M05, E07/M03, E07/M08). Detected in the three local sheep breeds to 527 with an average of 47.91 each primer combination, range 34 to 61, of which a total of 46 polymorphic bands accounted for 8.73% of the amplification of the total number of bands with a 4.18 average, each primer pair amplified polymorphic. That abundant genetic diversity of China's Xinjiang local sheep breeds. 3. According to the test results of 11 pairs of AFLP primer combinations calculated genetic similarity coefficient of Xinjiang three local sheep breeds, including of Kazakh sheep and Altay sheep genetic similarity coefficient (0.699), Kazakh sheep Bashibai sheep genetic similarity coefficient minimum (0.592), Altay sheep Bashibai sheep as (0.620). Higher genetic similarity between our local sheep breeds, the frequency of the genetic variation. 5. Using NTSYSpc Software UPGMA method, cluster analysis of China's Xinjiang three local sheep breeds, build a UPGMA dendrogram.
